Objectives:The research is to observe the effects of acupuncture and moxibustion on chronic unpredictable mild stress(CUMS) rats about the contents of 5-hydro-xytryptamine(5-HT), 5-hydroxyindole-acetic acid(5-HIAA), tryptophan(Trp), activity of 5-HT1A receptor in the prefrontal cortex tissue and their behaviors, to debate the effects of acupuncture and moxibustion on behaviors,5-HT system in prefrontal cortex of CUMS rats and their differences, to explore the antidepressant mechanism of acupuncture and moxibustion and their differences at some extent.Methods:45 male SD rats were divided into 2 groups randomly:normal group(containing 8 rats)and CUMS depression model building group("model building group" for short,containing 37 rats).Model building group was undergone chronic unpredictable mild stress for 2 weeks. After behavior tests, the successful models were divided into 4 groups randomly:model group, fluoxetine group, acupuncture group, moxibustion group. Each group contains 8 rats. All the treatment groups were intervened once a day,6 times per week and continued for 3 weeks. Fluoxetine group was given fluoxetine(1.8mg/kg/d) by gavage. Acupuncture and moxibustion group was given acupuncture and moxibustion the stimulation of "Baihui"(GV20) and "Dazhui"(GV14) for 20min/d relatively. CUMS continued during the treatment. The behavioral changes were determined by scores on open field test, percentages of sucrose consumption, body mass every week from beginning. All the groups were deprived of food and water for 12h after the final behavior tests, then they were killed and gotten prefrontal cortex tissue. The contents of 5-HT,5-H1AA, Trp were assayed by radioimmunoassay, and the relative binding affinities(constant and capacity) of 5-HT1A receptor were measured by radioligand binding assay and Scatchard plot analysis in a computer.Results:1.Behaviors:① In comparison with the normal group, open field test’s scores, percentages of sucrose consumption and body mass of model group significantly decrease(p<0.01).② Compared with model group, the horizontal and vertical scores on open field test of acupuncture, moxibustion, fluoxetine group were significantly increase after 1,2,3 week treatment relatively(p<0.01or0.05). Except open field test’s scores of moxibustion group was lower than acupuncture group on 21d(p<0.01or0.05), there are no obvious differences between them.③ Compared with model group, the percentages of sucrose consumption of all treatment groups increase significantly after 1 week treatment(p<0.01or0.05). Except the percentages of sucrose consumption of moxibustion group was lower than acupuncture group on 28d(p<0.05), there are no obvious differences between treatment groups.④ Compared with model group, the body mass of all the treatment groups increase significantly after 1 week treatment(p<0.01), and there aren’t any obvious differences between them. In comparison with themselves’ body mass on 21d, all the treatment groups increase significantly on 35d(p<0.01or0.05), but acupuncture group drop on 28d(p<0.01).2.5-HT system in prefrontal cortex tissue:① Compared with normal group, the content of 5-HT in model group, acupuncture group, moxibustion group significantly decrease(p<0.01 or0.05). The content of 5-HT in fluoxetine group is higher than model group(p<0.05), both acupuncture and moxibustion groups haveSno significant differences with model group. Moxibustion group is lower than fluoxetine group(p<0.01).②In comparison with normal group, the content of 5-HIAA in all treatment groups decrease(p<0.01 or0.05), there aren’t obvious differences between model group and normal group. Compared with model group, the content of 5-HIAA in all treatment groups decrease(p<0.01), acupuncture group is higher than moxibustion group(p<0.01or0.05).(3)In comparison with normal group, the content of Trp in model group decrease(p<0.01), fluoxetine group and moxibustion group significantly decrease(p<0.01 or0.05).Compared with model group, the content of Trp in all treatment groups decrease(p<0.01). There are no obvious differences between treatment groups.④ In comparison with normal group, the radio of 5-HT and 5-HIAA in model group decrease(p<0.01). Compared with model group, the radio of 5-HT and 5-HIAA in all treatment groups increase(p<0.01 or0.05), moxibustion group is lower than fluoxetine group(p<0.05).(5)In comparison with normal group, constant of high affinity 5-HT1A receptor in model group decrease(p<0.05), capacity isn’t have obvious difference. Compared with model group, constant and capacity of high affinity 5-HT1A receptor in all treatment groups have a rising trend, but without significant differences.⑥ In comparison with normal group, capacity of low affinity 5-HT1A receptor in model group decrease(p<0.05), constant isn’t have obvious difference. Compared with model group, constant and capacity of low affinity 5-HT1A receptor in all treatment groups have a rising trend, but without significant differences.Conclusions:1.Acupuncture and moxibustion on "Baihui"(GV20) and "Dazhui"(GV14) can significantly improve behaviors of CUMS rats and rise the body mass equally after 3 weeks’ treatment. But acupuncture and moxibustion have a different way clearly, the mechanism of them maybe different.2.Acupuncture and moxibustion on "Baihui"(GV20) and "Dazhu"(GV14) can significantly decrease the decomposition level of 5-HT in prefrontal cortex, rise utilization ratio of Trp and improve the less sensitive condition of 5-HT1A receptor in post synaptic membrane at some extent, their effects are equal basically.The antidepressant effects of acupuncture and moxibustion maybe related with their benign regulating to 5-HT system in prefrontal cortex.3.Law on curative effects of acupuncture and moxibustion on depression reflected by this experiment point out both of them have positive and identical meaning in treatment of depression.
