ObjectivesThis study based on the replenishing Qi resolving stasis and removing toxin compound formula,which belongs to the national TCM expert Professor Du Huaitang of Dongzhimen Hospital of Beijing University of Chinese Medicine.Through the model experiment, determine the time point of the model, set the prevention experiment, to explore the protective effect of replenishing Qi resolving stasis and removing toxin compound formula on the rats with ARDS due to LPS,and to observe the effect of the serum IL-1β, IL-6 and TNF-α levels.MethodsModel Search:18 male healthy SD rats were divided into 6 groups randomly, each group had 3 rats. According to animal husbandry standards, rats were adaptively fed for 1 week.2hgroup,4h group,6h group,16h group and 24h group single tail vein injection of lipopolysaccharide (LPS) solution (2mg/kg body weight), blank control group were not givenLPS, respectively at 2h,4h,6h,16h,24h after anesthesia executed.Take the lung tissue.Preventive experiment:50 male healthy SD rats were divided into 5 groups randomly, each group had 10 rats. According to the standard of animal feeding for 1 weeks. The low-dose group, middle-dose group high-dose group were given replenishing Qi resolving stasis and removing toxin compound formula Decoction low dose solution, replenishing Qi resolving stasis and removing toxin compound formula Decoction dose solution, replenishing Qi resolving stasis and removing toxin compound formulaDecoction high dose solution, model group received daily and volume of distilled water irrigation stomach in 7 days then creating model by vein injection of LPS (2mg/kg), the blank control group received same volume of distilled water by gavage for 7 days, without LPS injection. The 5 groups were executed afterl 6h anesthesia.Take the lung tissue and draw blood from abdominal aorta.ResultsModel exploration:a one-time tail vein injection of 2mg/ml LPS method of modeling,16 hours after the model can be.Prevention trials:lung pathology observation showed that the model group compared with the blank group, the pathological change significantly,compare with the model group, traditional Chinese medicine groups shows improvement in the alveoli and lung congestion in edema and inflammatory cell infiltration.Serum inflammatory factors:TNF-a:compared with the blank control group, model group’s the serum levels of TNF-a is higher(p<0.05). Compared with the model group, low dose of Chinese medicine group’s serum TNF -α does not decreased significantly (P> 0.05), themedium-dose group and high-dose group’s serum TNF-α levels decreased significantly (P< 0.001), but there is no significant difference (> 0.05) betweenmedium-dose group and high-dose group’s serum TNF-α levels. IL-1β: compared with the blank control group, model group’s serum IL-1β increased (P< 0.05). Compared with the model group, low-dose group’s serum IL-1 decreased (P< 0.05), medium-dose group and high-dose group’s serum levels of IL-1 P decreased significantly (P< 0.01); IL-6:compared with the blank control group, model group’s serum IL-6 increased (P< 0.05). Compared with the model group, low-dose group’s serum IL-6 was significantly decreased (P< 0.01) and medium-dose group’s serum IL-6 levels decreased significantly (P < 0.001), and high-dose group’s serum IL-6 levels decreased significantly (P< 0.01).ConclusionThemodelof2mg/mlLPSwasinjected through the tailvein, and themodelof acute respiratory distress syndrome could be induced after 16 hours. The replenishingQi resolvingstasisandremovingtoxincompoundformulacanreducethelunginjury,comparewiththem odelgroup,traditionalChinesemedicinegroupsshowsimprovement in thealveoli and lung congestion in edema and inflammatory cell infiltration.Theprotectiveeffect of replenishing Qi resolvingstasisandremovingtoxincompoundformulainduces acute respiratorydistresssyndrome may be related to reduced the level of TNF-α, IL-1β and IL-6 in serum of rats,whichisrelatedtothereduction of inflammatoryreaction.Inacertainrange,theeffect can be enhanced by increasingthedosageofreplenishing Qi resolvingstasisandremovingtoxincompoundformula.
