Purpose:To observe the effects of acupuncture on different acupoints of myocardial ischemia rats model of myocardial cells of ATP potassium channel (Kir6.1, Kir6.2) and compound protein (SUR2A, SUR2B) and protein kinase (PKA, PKG, PKCβ2) mRNA expression.The difference from myocardial cells ATP potassium channels and protein kinase gene expression, to investigate the expression level of the mechanism of electroacupuncture at Neiguan point on myocardial ischemia in the membrane ion channel gene.To reveal the meridian effect on the specificity of sequential transmission and influencing factors.Material and method:A total of 72 healthy male SPF SD rats were feed 7 days later(Ordinary feed and drinking water), the eighth day to choose 10 rats were random selected as the Contrast group model.Start experiment at 8:30 every morning of the remaining rats lasted two days.The 62 rats fell ill in the experiment and 22 rats died, survival rate was 64.5%, the remaining 40 mice were randomly divided into 4 groups(Model group, Neiguan group,Lieque group,Sham Acpoints group).A total of 5 groups with Contrast group, each group of 10. Clothed and Fixed rats when they are awake.The electro-acupuncture therapy on Neiguan group,Lieque group,Sham Acpoints group rats.Once a day,acupuncture therapy for seven days,used filiform needle into the 2~3mm,used dilatational wave,the frequency of 2~20Hz,the intensity of 2~3mm. Aacupuncture 20min.Don’t acupuncture with contrast group and model group.But at the same time and method of fetching and fixed Every day. Every group rats’ hearts are in the day after the acupuncture treatment at 8:30 in the morning to the experiment center of liaoning university of the experiment center.Put the heart tissues into the disinfection EP tube.Add 1ml the Trizol reagent and liquid nitrogen frozen.Stored in the -80 temperature.Used fluorescence quantitative PCR techniques.According to the instruction manual to amplification of target gene Kir6.1,Kir6.2,SUR2A,SUR2B,PKA,PKG,PKCP2 and GAPDH gens.In the relative of quantitative analysis,fluorescence quantitative PCR results is CT,the results to statistical processing.Results:1. The expression to each group of rats left ventricle cells Kir6.1. Kir62, SUR2A, SUR2B mRNA.Compared with contrast group:the increased expression of Model group,Neiguan group, Lieque group, Sham Acpoints group Kir6.1, Kir6.2, SUR2A, SUR2B mRNA (P<0.05). Compared with Model group:the down-regulated expression of Neiguan group, Lieque group (P<0.05). No significant expression of Sham Acpoints group (P>0.05). Compared with Neiguan group:the Increased expression of Lieque group,Sham Acpoints group (P<0.05). Compared with Lieque group:the Increased expression of Acpoints group (P<0.05).2.The Changes in the gene expression level of each rats’myocardial cell group.Compared with contrast group:the increased expression of Model group,Neiguan group, Lieque group, Sham Acpoints group PKA, PKG, PKCP2 mRNA (P<0.05). No significant expression of Sham Acpoints group (P>0.05). Compared with Neiguan group:the increased expression of Lieque group, Sham Acpoints group(P<0.05). Compared with Lieque group:the increased expression of Sham Acpoints group (P<0.05).Conclusion:1. Cupping in closed hole, column short hole of myocardial cell membrane ATP sensitive potassium channels Kir6.1, Kir6.2, SUR2A, SUR2B mRNA expression of related genes and protein kinase PKA, PKG, PKC beta 2) gene mRNA is significantly without going through the cave and the influence of the amount of the closed cavity, column short hole are against myocardial ischemia.2. Reduced clearance hole in myocardial cell membrane ATP sensitive potassium channels Kir6.1, Kir6.2, SUR2A, SUR2B mRNA expression of related genes and protein kinase PKA, PKG, PKC beta 2 mRNA related gene expression of the amount of work is better than the column short of acupuncture point, closed in the effect of acupuncture point against myocardial ischemia significantly.3.Electric acupuncture Nei Guan Xue of hand Jue Yin percardium can treatment of Dredge meridian4.In conclusion, Kir6.1,Kir6.2,SUR2A,SUR2BmRNA the expression changes has great effect to improvement of myocardial ischemia,the molecular mechanism of acupuncture on myocardial ischemia.This experiment further confirmed that acupuncture Significant points of the pericardium channel of hand-Jueyin has special dredge meridian for myocardial protection.
