Objective:1.To analyze the lower rate of postoperative recurrence metastasis of triple-negative breast cancer with Huaier aqueous treatment retrospectively. 2. To investigate the mechanisms that huaier surpress the invasion and metastasis in three triple negative breast cancer.Method:1. Statistics clinical cases, using statistical methods to verify the Huaier aqueous in reducing the rate of postoperative recurrence and metastasis of three triple negative breast cancer. 2. To verify the level of FN m RNA and protein expression of EGF-treated cells as analyzed by real-time PCR and western blotting in cell culture media. 3. To verify the levels of FN protein expression and m RNA expression in serum-starved cells pretreated with EGFR inhibitors, gefitinib or AG1478 for 30 min and then treated with 50 ng/ml EGF for 24 h, as analyzed by western blotting and real-time PCR. 4. To verify the levels of STAT3, Akt and Erk protein expression in serum-starved cells, pretreated wit u0126, LY294002 and Stattic, respectively, then treated with EGF for 15 min as analyzed by western blotting. 5. To verify the levels of FN protein and m RNA expression in cells pretreated with u O126, LY294002 and Stattic, respectively, then incubated with 50 ng/ml EGF for 24 h as analyzed by western blotting and real-time PCR, respectively. 6. To verify the levels of FN protein and m RNA expression in cells pretreated with Huaier aqueous then treated with EGF, as analyzed by western blotting and real-time PCR.7. To verify the levels of STAT3, Akt and Erk protein expression in cells pretreated with Huaier aqueous then treated with EGF, as analyzed by western blotting.Results:1. The rate of postoperative recurrence and metastasis of three triple negative breast cancer was surpressed by Huaier aqueous. 2. The level of FN expression is increased by EGF in a dose-dependent manner in breast cancer cells. 3. EGF-induced FN expression is reduced by EGFR inhibitors, gefitinib and AG1478, in MDA-MB-231 breast cancer cells. 4. Levels of STAT3, Akt and Erk protein expression was surpressed by inhibitor u0126, LY294002 and Stattic. 5. Levels of FN protein and m RNA expression was surpressed by inhibitor of u0126, LY294002 and Stattic. 6. Levels of FN protein and m RNA expression in cells pretreated with Huaier aqueous then treated with EGF. 7. Huaier aqueous suppresses EGF-induced FN expression through the inhibition of STAT3, Akt and Erk activity in MDA-MB-231 breast cancer cells.
