The high-performance liquid chromatography (HPLC) is one of the chromatographies, which has a good prospect, and the optimization of separation condition is the key in the separation of HPLC. Pharmacy field is one of the important fields to apply HPLC. No matter the analysis of Traditional Chinese Medicine components, or the content determination of compound preparation, or the research of pharmacokinetics, or the tracking analysis of pharmaceutical chemical, all need the use of HPLC. By using of the knowledge of mathematics, chemical statistics and others, the chromatographic workers have proposed a lot of theories and technique applying to optimizing the separation conditions. Computer technique, especially the development of computer programming language, makes it necessary and possible for the chromatographic workers to make use of it to settle the computing and optimizing problems.Using the visual program language-Visual Basic6.0(VB), I compiled the procedures of tetrahedron and simplex, and realized the automation of mobilexphase optimization. Both of the methods can be used in the isocratic elution of normal temprature in HPLC, and can be used to two, three and four members mobile phase.The tetrahedron method is proposed by Yu-qing Sun and his graduate student, which is used to optimize the strength and selectivity of mobile phase in the condition of isocratic elution. On the base of Wen Wu, I programmed the software by using of VB and the animated cartoon tool-3D. After finishing the software, I did the validated experiments using 8 aromatic compounds as sample.The simplex method is a classical method, which is able to optimize several factors simultaneously. To optimize by this method, the convergence velocity and the optimized result are due to the design of experiments to large extent. The common method to design the experiments is mixture-design technique. In my paper I used the "triangle" method, which is brought forward by Glajch, to design 5 turn experiments, then optimizing. I have also developed the procedure with tool of VB and Flash, realized the automotive optimization of simplex. Furthermore I studied the effect of the experiment's times and the object function to the optimizing process, improved them.
