Objective: This experiment taked two-month-old male SD rat as the object of this study.Through carry on the different intensity exercise to the big rat , this experiment had established the apoptosis model which laied in the big rat skeleton muscle cell. After the exercise, we instantly carried them into the high pressure oxygen warehouse to resume. Through study of the blood physiology biochemistry target and gene expression situation related to the apoptosis skeleton muscle cell of the control group, the exercise group, and HBO-restoration group, we tried to explain influence of hyperbaric oxygen restore to the different intensity exercise apoptosis cell and the related protein expression ,as well as explores its regulative mechanism sketchily.Methods: The control group (C, n=8), Incremental-loaded-training group (H, n=8), the exhaustive training group (GA, n=8), the hyperbaric oxygen restoration of incremental- loaded-training group (HC, n=8), the hyperbaric oxygen nexhaustive training group (GO, n=8). The Incremental-loaded-training group and the restoration group were trained for 8 weeks. Test methods: We observed change of skeletal muscle shape structural , through the light microscope and the electron microscope and determined Bcl-2 and p53 protein expression situation of muscle cell in the different category skeleton; Determined the MDA, SOD content in the different category skeleton of muscular tissue; Used the TUNEL to determine apoptosis skeleton muscle cell.Results: (1)Different load exercises made different influence for big rat skeleton muscle cell: 1st, the structure of skeletal muscle changes, the shrinkage of cell, the intercellular space had broadened,and the partial cell nucleus chromatin concentration approached model and so on ; 2nd, two traning groups had appeared the different degree of apoptosis. The incremental-loaded-training group's cell apoptosis degree and the sample masculine gender rate was higher than the exhaustive training group obviously. (2) 1st, Bcl-2 protein expression of each group of big rat's gastrocnemius had changed. The exercise training's two group of Bcl-2 protein expression obviously was lower than the control group. 2nd, p53 protein of each group of big rat's gastrocnemius expression had changed. The two group of exercise training's p53 protein expression obviously were higher than the control groups. 3rd, Bcl-2 protein expression of the hyperbaric oxygen restration group had raised than the control group's.BUT the p53 protein expression of the hyperbaric oxygen restoration group were dropping. Each group of exercise group's MDA and SOD content, as well as in the blood ammonia, the hemoglobin content had change correspondly.Conclusion: (1) The different exercise caused different apoptosis degree in the big rat skeleton muscle cell, which explained physiological exercise could influence apoptosis degree, namely the harder exercise were, the weaker apoptosis degree were .(2) Hyperbaric oxygen could improve the environment of cell, and eliminated the oxygen free radical content, then prevente the oxygen free radical to induce the possible way of apoptosis cell. (3) The different intensity's exercise made different influence of the related gene expression, but the hyperbaric oxygen had the remarkable influence.The hyperbaric oxygen had the remarkable enhancement to the Bcl-2 gene expression, but p53 protein masculine gender rate had obviously reduced, which could explant hyperbaric oxygen has the remarkable effect with eliminating the apoptosis of big rat skeleton muscle cell.
