In modern society, with the rapid development of the communication and network of technology, Distance Education (DE) has been unprecedented attention and development, become an important component of the system in the system of higher education and lifelong education, and showed self-learning and tele-training trend. The quality is the lifeline of DE. This paper concerns the importance of the quality of e-courses.The sociality of human learning and "space-time separation of teachers and students" of DE decided that important work of DE is building more species diversity of interactive activities. Effective interactive teaching and learning can improve the quality of DE. Interaction always is the focus and hot issue of DE. This paper describes learning theoretical basis of the interaction, comparatively analyze the interaction of modern DE with the traditional classroom teaching and traditional DE, study classification and levels of interaction of DE, point out that interaction is the important indicators to measure quality of e-courses. Finally, we analyze limitations of Interaction of the present stage of DE practice: media technology development and application constraints; Design and Development of Interaction positioning deviation; Social interaction lack; Distance learner's physical location on the effects of interaction.This paper focuses on distance learners and the learning process from the perception of interaction, examines the applications of Agent technology in DE, use Microsoft Agent to create interface agent characters and enhance interaction based on the design and development of specific types of language e-course. During design and development of the an English e-courses, according to different learning content needs, we called Ms Agent for the three movie characters (played respectively learning partner, teacher, course manager role) and applied to the parts of thee-courses, such as interface welcome page, extended pages, introduction of e-courses, the learning courses, writing- listening exercises. We establish vivid language learning environment, rich content form and interactive interface,increase social interaction in an attempt to reduce the learning mental load of machine-oriented learner, so as to enhance the learning interest and maintain effective behavior of distance learning.This paper based on the specific work of the practice. On the basis of practice, we tried to sum up the relevant experience about design and development of e-courses' interaction used role of Ms agent: Appropriate creation of interface agent character based on learners'traits and learning contents as the practical needs; The design details (such as motor, language, facial expressions, etc.) of characters to respect the learners and follow interpersonal communication at the most basic courtesy and friendly, also ask for the characters to reflect distinct personality; Provide Hide order to enhance controllability of learners to interface agents and so on. These experiences have the general references to the use of Ms Agent that can improve interface operation's interaction of e-courses.
