Ezrin is a member of the widely distributed ezrin-radixin-moesin (ERM) family, which is a cross-linker of the actin cytoskeleton and plasma membrane , and play a key role in cell morphous keeping and signaling pathways regulation. our previous studies showed that the upregulated expression of Ezrin might be correlated with the malignant transformation of normal esophageal squamous cells to cancer cells, and RNAi analysis revealed that ezrin could inhibit the ability of proliferation and invasiveness of ESCC cells effectively. yet the clinical research showed that Ezrin was associated with poor survival of patient with ESCC and likely act as a marker for the prognosis.Followed above research, we plan to study the ezrin molecular mechanisms in migration including the mutation in T567 of Ezrin.First, by transfection the plasmids including pEGFP-N1-ezrin-T/A(T567A), pEGFP-N1-ezrin-T/D(T567D), pEGFP-N1-ezrin-Nter(N terminal, 309aa), we got the cells which be used to detected the abilities of the migration and proliferation. The result showed that the cell transfected pEGFP-N1-ezrin-T/D appear the more movement than pEGFP-N1's; and pEGFP-N1-ezrin-T/A and pEGFP-N1-ezrin-wt groups have no changes. However, the pEGFP-N1-ezrin-Nter group showed predominance decrease. To sum up, the ezrin T567 play the important role in facilitating the cell migration.Second, the detection of above cell proliferation showed that pEGFP-N1-ezrin-T/D group cell have more stronger capability of the proliferation while the others appear no changes.Third, we assay the cell cycle of above cells using flow cytometer. The result showed that pEGFP-N1-ezrin-T/A and pEGFP-N1-ezrin-Nter group cells were inhibited in G1 phase, and pEGFP-N1-ezrin- wt group cell was inhibited in S phase, pEGFP-N1-ezrin-T/D group cells were inhibited in G2/M phase.To collected together, ezrin possess the potential to increase the cell migration especially the T567 modification, and may be associated the phosphorylation process.
