Objective:In this research,we constructed and identificated plasmid vector of microRNA on Pneumocystis carinii major surface glycoprotein gene's upstream conserved sequence.Methods: microRNA oligonucleotide targeting PC MSG UCS gene was chemically synthesized and was inserted into microRNA-Ready plasmid vector pcDNA?6.2-GW/EmGFPmiR. The recombinant plasmid, pPC-UCSm, were dentified by gel electrophoresis and DNA sequencing.Results: Gel electrophoresis and DNA sequencing showed that the recombinant plasmid containing the correct and full microRNA oligonucleotide.Conclusion: The microRNA plasmid, pPC-UCSm, was constructed successfully.
