Objective:To investigate the effects of baicalin on proliferation and the expression of transforming growth factor beta 1(TGF-β1) of rabbit Tenon's capsule fibroblasts.Methods:MTT assay was used to examine the effect of baicalin with different concentration(15,30,45,60μg/ml) on the proliferation of the fibroblasts in 24,48,72 hours ,while the cell cycle was analyzed by FCM.The effect of tranilast on the expression of TGF-β1 mRNA was determined by reverse transcriptase-polymerase chain reaction(RT-PCR) analysis. The experiment of FCM and RT-PCR include Baicalin 30μg/ml group and contrast group.Results:1. Primary culture of rabbit tenon's capule fibrobiasts,the cells emigrated from the tissue on the sixth day and covered in a whirlpool like on the 14th day.2. In 15 60μg / ml with the concentration,Baicalin inhibited the proliferation of fibroblasts and the effect were in a time-and-concentration dependent manner.3.FCM revealed that fibroblasts population at the S phase were significantly decreased from 35.1733%±1.8441% to 22.1333%±0.6901% after the baicalin treatment(P<0.01).4.RT-PCR showed that baicalin may downregulated the expression of TGF-β1 mRNA of rabbit tenon's capsule fibroblasts.The gray values decreased from 1.1502±0.0285 to 1.0162±0.0028(P<0.01).Conclusion : The findings suggested that baicalin could downregulated the expression of TGF-β1 mRNA and inhibited the proliferation of fibroblasts.
