Objective: Observing the hypocrellin-mediated LED photodynamic therapy apoptosis on keloid fibroblasts of nude mice.Methods: Observing the differences of proliferation and apoptosis between keloid fibroblasts and normal skin fibroblasts with HE staining,proliferating cell nuclear antigen (PCNA) and in situ end labeling (TUNEL).After human keloid tissues have been transplanted on nude mice and successfully established Keloid nude model, hypocrellin-mediated LED photodynamic therapy experimental group, LED light control group, photosensitizer hypocrellin control group and control group have been set up. Detecting the proliferation and apoptosis of keloid fibroblasts after hypocrellin-mediated LED Photodynamic therapy with PCNA and TUNEL.Results: Compared with normal skin, keloids showed thick bundles of parallel or spiral arrangement of collagen fibers and significantly increased fibroblasts in HE staining, and PCNA test showed the significantly increased proliferation of keloid fibroblasts while TUNEL test showed the significantly reduced apoptosis of keloid fibroblasts. Hypocrellin-mediated LED photodynamic therapy have significantly inhibited the proliferation of keloid fibroblasts and induced apoptosis, and compared whit the control group there was significant difference(P<0.01). Hypocrellin-mediated LED photodynamic therapy appeared more effective than single-factor treatment as LED light or hypocrellin, compared with LED light control group and photosensitizer hypocrellin control group, there were significant differences(P<0.01). LED light treatment or hypocrellin treatment can inhibite the proliferation and induce the apoptosis of keloid fibroblasts to some extent, but the comparison between the two treatments has no significant difference(P>0.05).Conclusion: Hypocrellin-mediated LED photodynamic therapy have significantly inhibited the proliferation and induced apoptosis of keloid fibroblasts on nude mice model, and a new treatment for Keloid could be found.
