Objective:Investigate the protective effect of Salviamiltiorriza Bunge in vitro liver cold preservation and affect of HO-1 expression at different save time, and identify protective effect on the liver and its possible mechanism.Methods:1.72 SD male rats were divided into 3 groups randomly:group A: lactic acid Ringer's solution (LR) infusion group, n=24; group B:Salviamiltiorriza Bunge (Salviamiltiorriza Bunge, SB) 300mg/Kg+Ringer lactate solution infusion group (SB+LR), n=24; group C:SB+LR infusion group, n=24. Group A and group B with 5ml normal saline intraperitoneally 24 hours before modeling. Group C ZnPP (1.5mg/Kg), intraperitoneal injection 24 hours before modeling.Each group before the infusion, respectively, save 1 hour,3 hours and 6 hours, stored for different placement. Specimens were cut at different times by each group divided into 4 subgroups, each sub-group of 6 rats.2. Observed morphous of hepatocyte and hepatic sinusoid endothelial cell by light microscope after the liver preserved in different time of each group; 3, Observed morphous of hepatocyte and hepatic sinusoid endothelial cell by electron microscopy after the liver preserved in different time of each group; 4, Detect mitochondrial Ca2+-ATP activity and mitochondrial Ca2 concentration in different time of each group 5, Detect HO-1mRNA expression detected by reverse transcription polymerase chain reaction (RT-PCR) after the liver preserved in time compartments of each group.Results:1, Liver biopsy specimens showed group B liver cells, liver sinusoidal damage was light than the group A, and group C liver cells, liver sinusoidal damage damage serious than the other two groups; 2, Electron microscopy showed that group B liver mitochondrial damage significantly lighter than the group A, group C liver mitochondrial damage serious than the other two groups; 3 Group B liver samples of mitochondrial Ca2+-ATP activity and mitochondrial Ca2+levels higher than the group A of each time, while the group C of mitochondrial Ca2+-ATP activity and mitochondrial Ca2+content significantly lower than the other two groups.4,RT-PCR results showed that, group B HO-1mRNA is upregulated than group A, while group C was significantly reduced;Conclusion:Salviamiltiorriza Bunge can induce HO-1 in mRNA, this expression may be its liver protective effects in vitro one of the mechanisms.
