Objective:To understand research status about Daphne genkwa by consulting literatures published in home and abroad. System research on chemical constituents of,in order to get more control substance of plant drug,providing reference substance for quality standard. Further research on quality standards of Daphne genkwa medicinal materials medicinal,which provide reliable data for relevant standards of the Chinese Pharmacopoeia 2010 edition.Deepen the understanding of Daphne genkwa, but also lay a foundation for its further development and utilization.Methods: This paper consists of four parts,Part I:systematic study of the historical evolution of the Daphne genkwa, chemical constituents, pharmacological effects, toxicity and quality control. Part II: Reflux extraction with 80% ethanol,obtained a variety of 11 compounds by the method of column chromatography, identifying its structure through a variety of spectral data and physical and chemical properties.Part III:Research of genkwanin and yuanhuacine as reference substance in daphne genkwa medicinal materials were increased;The genkwanin,yuanhuacine and reference crude drug for contrast by TLC was increased under differential item;The limit of alcohol-soluble extractive was increased under the extract item;Using genkwaninand yuanhuacine as reference substance for the control of the HPLC assay method and the limits of requirements were increased under determination item.Part IV: Conclusion.Results:11 compounds were isolated from daphne genkwa,and were identified separately as:genkwanin(1),apigenin(2),luteolin(3),luteolin-7-O-β-D-glucoside(4),apigenin-4',7-dimethyl ether(5),daphnoretin(6),β-sitosterol(7),daucosterol(8),daphnoretin-7-O-β-D-glucopyranoside(9) lindenglycosides (kaempferol-3-β-D-(6-O-trans-p-coumaroyl) glucopyranoside 10) (quercetin 11).Among them, compound 4 9 and 11 was isolated from the daphne genkwa for the first time.The preparation process of genkwanin and yuanhuacine were established;Using genkwanin and yuanhuacine as reference substance for the TLC, and developing solvent was : toluene - ethyl acetate - formic acid (8:4:0.2 ); the content of genkwanin in daphne genkwa medicine shall not be less than 0.3%, yuanhuacine should be controlled between 0.002% ~ 0.06%.Conclusion:In this paper, the reference substance which isolated from Daphne genkwa not only provide certified reference material for further control quality of Daphne genkwa, but also provide certified reference material for identifying fingerprint of Daphne genkwa as much as possible.The quality standard uesd genkwanin as reference substance for the TLC, determination methods and the limits of request has been received by the Chinese Pharmacopoeia 2010 edition; The established quality standards improved the level of quality control of Daphne genkwa, and providing a scientific basis.for the clinical safety and efficacy of drug use.
