Objectives:This study was designed to investigate the expression and relationship of B7-H4 and FOXP3 protein in normal breast and benign lesional and breast cancer, and their correlation with clincopathologic features, for provide evidence for breast cancer of diagnosis and prognostic evaluation.Methods:283 paraffin-embedded specimens were collected , including 267 cases of invasive breast cancer and 15 cases of benign lesions; 11 cases of fresh normal breast,10 cases of fresh benign lesions and 10 cases of fresh invasive breast cancer were collected; The expression of B7-H4 and FOXP3 was measured by immunohistochemistry SP methods in 11 cases of normal breast,25 cases of benign lesions and 277 cases of invasive breast cancer; The expression of B7-H4 mRNA was measured by in situ hybridization in 10 cases of normal breast,10cases of benign lesions and 20 cases of invasive breast cancer; Analyze the relationship between the expression of B7-H4 and FOXP3 and the pathological characteristics; Analyze the relationship between the expression of B7-H4 mRNA and the pathological characteristics.Results:1. The positive rate of the expression of B7-H4 in breast cancer (88.44%) is higher than that in normal breast (9.09%) and benign lesions(44.00%), the difference has statistical significance(P<0.0001 and P<0.0001). And the positive rate of the expression of B7-H4 in benign lesions is higher than that in normal breast , the difference has statistical significance(P<0.0001).2. The positive rate of the expression of FOXP3 in breast cancer (86.64%) is higher than that in normal breast (27.27%) and benign lesions(36.00%), the difference has statistical significance(P<0.0001 and P<0.0001). There is no statistical significance on positive rate of the expression of FOXP3 between normal breast and benign lesions(P>0.05).3. There is no statistical significance between The positive rate of the expression of B7-H4 and age,tumour size,histological classification(P>0.05). The positive rate of the expression of B7-H4 in histological gradeâ… is lower than that in histological gradeâ…¡and histological gradeâ…¢, the difference has statistical significance(P=0.025, p=0.020). There is no statistical significance on positive rate of the expression of B7-H4 between histological gradeâ…¡and histological gradeâ…¢(P>0.05). The positive rate of the expression of B7-H4 in the lymph node matastasis group is higher than that in the no lymph node matastasis group , the difference has statistical significance(P=0.009).4. There is no statistical significance between The positive rate of the expression of FOXP3 and age,tumour size,histological classification(P>0.05). The positive rate of the expression of FOXP3 in histological gradeâ… is lower than that in histological gradeâ…¡and histological gradeâ…¢, the difference has statistical significance(P=0.003 and P=0.022). There is no statistical significance on positive rate of the expression of FOXP3 between histological gradeâ…¡and histological gradeâ…¢(P>0.05). The positive rate of the expression of FOXP3 in the lymph node matastasis group is higher than that in the no lymph node matastasis group , the difference has statistical significance(P<0.0001).5. B7-H4 is positive correlation with FOXP3(r=0.306,P<0.001)6. Experssion of B7-H4 mRNA was obseverd in the cell cytoplasm. The positive rate of the expression of B7-H4 mRNA in normal breast group , benign lesions group and breast cancer group is 90%, 80% and 90% respectively. There is no statistical significance on positive rate of the expression of B7-H4 mRNA among that in normal breast group , benign lesions group and breast cancer group (P>0.05).Conclusions:1. There were different expression of B7-H4 and FOXP3 in normal breast,benign lesions and breast cancer.2. There is no statistical significance between the positive rate of the expression of B7-H4 and FOXP3 and age,tumour size,histological classification, however, there is statistical significance between The positive rate of the expression of B7-H4 and histological grade,Lymph node matastasis.3. B7-H4 is positive correlation with FOXP3.
