OBJECTIVE: To investigate the effects of osteoprotegerin (OPG) on apoptosis in cultured aorta vascular smooth musle cell(VSMC) of sprague dawleyd rat and its possible mechanismsMETHODS : 1.The VSMC of rat was treated with different concentration of OPG for 24 hours, the rate of apoptosis of VSMC was observed by flow cytometry(FCM) and TdT-mediated dUTP-biotin nick end labeling(TUNEL). 2.The VSMC of rat was treated with OPG(20 ng/ml) and 200 ng/ml TRAIL or anti-TRAIL(100 ng/ml)for 24 hours,then the rate of apoptosis of VSMC was observed by FCM and TUNEL.RESULTS: 1.The low concentration of OPG(0-20 ng/ml)could induce the apoptosis of VSMC in vitro with a concentration-dependent fashion. When the concentrateion of OPG achieved 20ng/L, the effect of induction on the apoptosis of VSMC was highest . 2. The high concentration of OPG(50-400 ng/ml)can deduce the rate of the apoptosis of VSMC in vitro in a concentration-dependent fashion. When the concentrateion of OPG achieved 200ngmol/L, the effect of inhibition on the apoptosis of VSMC was highest. 3. In the condition of no serum, there are no distinction of apoptosis with the concentration of 20ng/ml of OPG wherther or not 200 ng/ml TRAIL or 100 ng/ml anti-TRAIL.CONCLUSION: The low concentration of OPG could induced the rate of apoptosis VSMC with a concentration-dependent fashion, The high concentration of OPG could deduce the rate of apoptosis VSMC also with a concentration- dependent fashion. The apoptosis mechanisms of VSMC may not be related to TRAIL.
