Objective To discuss the prevention and treatment effect of the matrine to rat hepatocirrhosis,which induced by carbon tetrachloride (CCL4) and alcohol. Establishing the intervention effect of TGF-βand HGF in liver fibrosis and different concentrations of matrine on liver fibrosis in rat liver tissue TGF-βand HGF expression, proposed for the clinical treatment of hepatocirrhosis in experimental basis.Method 52 male Sprague-Dawley rats were randomly divided into 5 groups:blank control group,model group,γ-interferon group,matrine low-dose group and matrine high-dose group. Model group,γ-interferon group,matrine high-dose group and matrine low-dose group were prepared by carbon tetrachloride (CCL4) subcutaneous injection and alcohol-fed to get rat liver fibrosis model,and the blank control group treated with the same volume of normal saline solution to fill the stomach,every group were medicated by the above means for 8Ws constantly. Rats in each group harvested after modeling 4W,8W,blank control group were sacrificed 2 rats,12 rats were randomly killed in other groups,total of 26 rats. Using RT-PCR and Western Blot to detect TGF-β1 /HGF mRNA and protein expression of each group. Also used optical microscope by means of HE dyeing to observe the level of rat liver tissue fibrosis .Results 1,The establishment of rat model of hepatic fibrosis:The sss of Hepatic fibrosis histologically in the model group was higher than the blank control group,the blank control group:x±s=0;the model group:x±s=11.619±1.563,(P>0.05).The have no difference between low-dose group of matrine group ( x±s=10.8500±1.365)and the blank control group(P>0.05).Matrine high-dose group: x±s=7.4287±0.694;interferon group : x±s=7.5851±0.798 ; the two group lower than the model group(P>0.05). the high-dose group of matrine have no significant difference with γ-interferon group(P>0.05)2,The effects of matrine on TGF-β1 in the rat's hepatic fibrosis tissue:TGF-β1 was positive In the blank control group. Compared with normal Liver tissue the fibrosis liver tissue levels of TGF-β1 significantly increased (P<0.01), the expression of TGF-β1 in matrine high-dose group andγ-interferon group were both lower compared to model group (P<0.05), and high-dose group of matrine TGF-β1 was the lowest. And the longer use of the matrine, the more reduce of the TGF-β1 . The expressiong of TGF-β1 in matrine low-dose group was lower than model group compared to liver tissue expression of , but have no significant difference (P>0.05).Sophora flavescens ait. The have no significant difference with matrine high-dose group andγ-interferon group (P > 0.05).3,The Effect of matrine on HGF in hepatic fibrosis of rat liver tissue:The expression of HGF in liver tissue in model group compared with blank control group was significantly lower (P <0.01). Compared with model group, the expression of HGF in matrine high-dose group and theγ-interferon group have increased (P <0.05), the high-dose group of matrine HGF increased the most obviously .Matrine high-dose group andγ-interferon group can increase the content of HGF in liver tissue, and with the extension of time of using matrine, HGF in high-dose group andγ-interferon group elevated more obviously.Conclusion:1.rat model induced by carbon tetrachloride (CCL4)for 8Ws was adopted by characteristics of a typical animal model of liver injury.2.The mechanisms of matrine on HF may be inhibit the activity of TGF-β1 and adjust the level of HGF .
