Objective:The aim of the study is to explore whether mucosolvan can increase levofloxacin clear Staphylococcus epidermidis biofilm infection on the orthopaedic implants by combined mucosolvan with levofloxacin to treat rat knee joint prosthesis infection modelMethods:1: Experimental research in vitro that Mucosolvan influence on Staphylococcus epidermidis biofilmTesting the formation capability of Staphylococcus epidermidis(ATCC35984),MIC of levofloxacin to Staphylococcus epidermidis(ATCC35984). Observing the effect of Mucosolvan to Staphylococcus epidermidis extracellular mucilage (ESS); Experiments in vitro study biofilm morphology that Mucosolvan influence on Staphylococcus epidermidis biofilm by fluorescence microscopy, laser scanning confocal microscopy.2: Experimental research in vivo that Mucosolvan influence on Staphylococcus epidermidis biofilmEstablished model of knee prosthesis by SD rats, vaccinating Staphylococcus epidermidis (ATCC35984) to articular cavity two weeks later after operation, creating arthrosis prosthesis infection model. injecting Inj Mucosolvan or Levofloxacin to articular cavity three weeks later, observing animal body,WBC number,biolfilm bacteria count on prosthesis surface,The tissues germiculture around joints,fluorescence microscope and the observation of bacterial biofilm morphology by laser scanning confocal microscope etc.; observing the prevention and control efficacy of Mucosolvan to Staphylococcus aureus infection in prosthetic articular. Results:1,Staphylococcus epidermidis(ATCC35984)have good ability of forming biolfilm; Mucosolvan can decrease extracellular mucilage (ESS) from the process of biolfilm formation(ATCC35984); Levofloxacin combined effect of ambroxol on Staphylococcus epidermidis, reduce the number of bacteria.2,in vivo experiment showed that ambroxol knee joint in rats infected with levofloxacin reduced the number of viable cells, ESS , etc.Conclusion:1,Experiments in vitro proved Mucosolvan can decrease Staphylococcus epidermidis from producing ESS, the combination of mucosolvan and levofloxacin have synergism in inhibiting Staphylococcus epidermidis from forming biofilm.2,Experiments in vivo proved Mucosolvan can enhance levofloxacin clear Staphylococcus epidermidis in biofilm.
