Objective:Hyperlipidemia(HLP) is due to confusion of synthesis and cleanup of lipoproteins in the blood. The plasma lipoprotein is abnormal and the blood lipid is heighten.It belongs to "phlegm"and"blood stasis"in the traditinal Chinese medicine. Essential hypertension (hypertension) is a kind of systemic inflammatory disease, which is characterized by arterial blood pressure increased and can be accompanied by functional or organic changes of the heart, blood vessels, brain and kidneys and other organs; it is belong to the category of traditional Chinese medicine "Vertigo" " headache ", both of them are common clinical disease, and on the rise. In this study, we tried to establish the hyperlipidemia model and the hypertension model in mice, respectively, to investigate the suppressive effects of Rhizoma Alismatis decoction and its ingredients on lipid metabolism and hypertension in mice. In addition, glucose metabolism was observed on normal mice. Methods:1 To develop a new method for establishment of hyperlipidemia animal model by comparing different methods in mice. Four groups of mice were respectively given①normal diet,②intragastric administration of fat emulsion once a day,③high fat diet whole day and④high fat diet whole day with giving intragastric administration of fat emulsion every two days. The blood pressure and serum levels of TC, TG and HDL-C were measured at the first day, the seventh day, the fourteenth day and the twenty-first day. At the end of experimental period, the organ coefficient was calculated.2 Mice were fed high fat diet to set up the model of hyperlipidemia. Mice were randomly divided into 6 groups:blank control, model, positive control, Rhizoma Alismatis, Rhizoma Atractylodis Macrocephalae and compound prescription, Intragastric administration was given continuously for 4 weeks,body weight were recorded weekly. The content of TC, TG, HDL-C and activities of ALT and AST in serum were measured respectively. The organ coefficient was also calculated.3 The model of hypertension was set up by hypodermic injection of testosterone propionate. Mice were randomly divided into 5 groups: model, low-dose Rhizoma Alismatis, high-dose Rhizoma Alismatis, Rhizoma Atractylodis Macrocephalae and compound prescription (Rhizoma Alismatis decoction).After 16 h of fasting, mice were given corresponding samples orally, The BP was measured before and 1,2, 4,6, and 8 h after the administration.The normal mice were divided into three groups:one group was the control and the others, the low-dose and high-dose Rhizoma Alismatis.After fasting for 18 h, mice were given 2g/Kg glucose solution as control and 2g/Kg glucose containing 20g/Kg and 30g/Kg Rhizoma Alismatis via a gastric tube, respectively.The blood was collected from the tail vein before (0) and 30,60, and 120 min after administration, and plasma glucose was calculated.Result:1 The establishment of the hyperlipidemia model in mice Compared with other groups, the liver coefficient of the group with high fat diet whole day and intragastric administration of fat emulsion was statistically increased, the blood pressure level of this group statistically increased after treatment of fourteen days (P<0.01); The content of TC and TG statistically increased after treatment of seven days and fourteen days, respectively (P<0.01); The content of HDL-C decreased after treatment of seven days (P<0.01); By comparison with the fourteenth day treatment, the blood pressure levels and the high blood lipids level of the twenty-first day treatment stay steadily. The group with high fat diet whole day and intragastric administration of fat emulsion had low mice morality.2 The effects of Rhizoma Alismatis decoction and its ingredients on experimental hyperlipidemiaCompared with the model group, the Rhizoma Alismatis and compound prescription markedly decreased the TC,TG and increased the HDL-C in serum, the activities of ALT and AST significantly decreased in serum; Rhizoma Alismatis, Rhizoma Atractylodis Macrocephalae and compound prescription decreased the organ coefficient of the liver and spleen in a certain degree; Compared with the Rhizoma Alismatis group, the compound prescription not only prevented obesity development, but also caused a rapid decrease in the organ coefficient of the liver after intragastric administration for 4 weeks.3 The effects of Rhizoma Alismatis decoction and its ingredients on experimental hypertensionCompared with the model group, both the Rhizoma Alismatis and compound prescription can decreased the SBP to some extent, but Rhizoma Atractylodis Macrocephalae almost didn't change the SBP. Meanwhile, Rhizoma Alismatis had a more significant effect than compound prescription in decreasing the SBP. In addition, Rhizoma Alismatis can lower plasma glucose in normal mice.Conclusion:1 The model of hyperlipemia in mice is successful.2 The compound prescription is reasonable, Rhizoma Atractylodis acts in synergy with Macrocephalae Rhizoma Alismatis for lowering hyperlipidemia.3 Rhizoma Alismatis combined with Rhizoma Atractylodis Macroce phalae could lower hypertension in synergy.Rhizoma Alismatis could act the pole of improving glucose metabolism and regulating glucose tolerance,on some extent,its effects were dose-dependent.SUBJECT WORDS:Rhizoma Alismatis decoction; hyperlipidemia; hypertension; plasma glucose; synergistic effect...
