Ec9706 cells were cultured in the environment of 37℃,5%CO2 with routine method. The logarithmically growing Ec9706 cells were used.2.1 Different groups of medicine:⑴control group: isotonic Na chloride;⑵groups of artesunate(AS): 0.01,0.1,1.0μmol/L;⑶groups of adriamycin (ADM): 0.02,0.2,2.0ng/μl;⑷groups of artesumate(AS) combined with adriamycin(ADM):①AS 0.01μmol/L+ADM 0.02,0.2,2.0ng/μl;②AS 0.1μmol/L+ ADM 0.02,0.2,2.0ng/μl;③AS 1.0μmol/L+ ADM 0.02,0.2,2.0 ng/μl.2.2 The examining of different groups of Ec9706 cell by flow cytometry(FCM). 2.3 The expression of ABCG2 protein in Ec9706 cell by RT-PCR.Result:1 The expression of ABCG2 protein in different esophageal tissues by IHC and FCM1.1 The expression of ABCG2 protein in 60 cases of esophageal carcinoma,60 cases of atypical hyperplasia tissue and 60 cases of normal mucosa tissue were detected by IHC, the results showed that the positive expression rate of ABCG2 protein (83.33%) in esophageal carcinoma was higher than those in atypical hyperplasia tissue (31.67%) and normal mucosa tissue (3.33%)(P<0.01).The expression of ABCG2 protein was not correlated with age and sex(P>0.05),but the expression was correlated with cell differential degree, infiltrate depth and lymph node metastasis(P<0.05).1.2 The expression of ABCG2 protein in 60 cases of esophageal carcinoma,60 cases of atypical hyperplasia tissue and 60 cases of normal mucosa tissue were detected by FCM, the results showed that the ABCG2 protein content (Fluorescence Index, FI) in esophageal carcinoma was higher than that in atypical hyperplasia epithelium and normal mucosa (P<0.01).2 The expression of ABCB1 protein in different esophageal tissues by IHC and FCM2.1 The expression of ABCB1 protein in 60 cases of esophageal carcinoma,60 cases of atypical hyperplasia tissue and 60 cases of normal mucosa tissue were detected by IHC, the results showed that the positive expression rate of ABCB1 protein (78.33%)in esophageal carcinoma was higher than those in atypical hyperplasia tissue (36.67%) and normal mucosa tissue (6.67%)(P<0.01).The expression of ABCB1 protein was not correlated with age and sex(P>0.05),but the expression was correlated with cell differential degree, infiltrate depth and lymph node metastasis(P<0.05).2.2 The expression of ABCB1 protein in 60 cases of esophageal carcinoma,60 cases of atypical hyperplasia tissue and 60 cases of normal mucosa tissue were detected by FCM, the results showed that the ABCB1 protein content (Fluscence Index, FI) in esophageal carcinoma was higher than that in atypical hyperplasia epithelium and normal mucosa (P<0.01).3 The expression of LRP protein in 80 cases of esophageal carcinoma and 80 cases of normal mucosa tissue were detected by IHC, the results showed that the positive expression rate of LRP protein (55.00%)in esophageal carcinoma was higher than those in normal mucosa tissue (35.00%)(P<0.01).The expression of LRP protein was not correlated with cell differential degree, infiltrate depth and lymph node metastasis(P>0.05).4 The apoptosis rates of Ec9706 cell were effected by ADM,AS and ADM combined with AS. The apoptosis rates of Ec9706 cell of ADM used alone for 24h were higher than those in the control group(P<0.05). The apoptosis rates of Ec9706 cell of AS used alone for 24h were no significance than those in the control group(P>0.05).The apoptosis rates of Ec9706 cell of AS combined with different concentrations of ADM were higher than those in groups of ADM used alone(P<0.01).5 The expression of ABCG2mRNA in Ec9706 cell by RT-PCR and the expression of ABCG2mRNA in Ec9706 cell of AS used alone for 24h,the result showed that the expression of ABCG2mRNA in Ec9706 cell of AS used alone was lower than those in Ec9706 cell (P<0.01).Conclusion:1 The expression of ABCG2,ABCB1 in esophageal carcinoma were significantly higher than normal mucosa and the expression was correlated with cell differential degree, infiltrate depth and lymph node metastasis. The result cued that hyper express of ABCG2,ABCB1 in esophageal carcinoma maybe participate in carcinogenesis of esophageal carcinoma andproduction of drug fast.ABCG2 maybe become a new reversal target ofdrug fast.2 The expression of LRP in esophageal carcinoma was significantly higher than normal mucosa and the expression was not correlated with celldifferential degree, infiltrate depth and lymph node metastasis.The result cued that hyper-express of LRP in esophageal carcinoma maybe participate in production of drug fast. 3 The apoptosis rates of AS combined with different concentrations of ADM were higher than those in groups of ADM used alone, the result cued that both of AS and ADM have synergy to resist esophageal carcinoma.4 The apoptosis rates of Ec9706 cell of AS used alone for 24h were no significance than those in other groups, but the expression of ABCG2mRNA in Ec9706 cell of AS used alone was lower than those in Ec9706 cell ,the result cued that the expression of ABCG2mRNA in Ec9706 cell were reduced by AS, then the expression of ABCG2 protein were reduce, so the concentration of anticancer drugs is higher than others. | | Keywords/Search Tags: | ABCG2, ABCB1, LRP, Artesunate(AS), multidrug, resistance(MDR), immunohistochemistry(IHC), Flow Cytometry(FCM) | PDF Full Text Request | Related items |
