Abstract:Objective:Asthma is common and frequently encountered disease, which affect human health and endanger human life. The uteroglobin related protein 1 (UGRP1) has closely relation with asthma according to recent research, but the change rule of UGRP1 and its mRNA still has dispute. This experiment observe the change rule of UGRP1 and the effect of dexamethasone (DM) in successive time (6h and 24h), furtherly disclose the function of UGRP1, explain the pathogenesis of asthma and find new theory which help to treat this disease. Methods:Divided mouse into three groups randomly, asthma group(A),sensitized control group (K),dexamethasone intervened group(D).In asthma groups Mouse (6 wk old) were sensitized by an intraperitoneal injection of 50μg ovalbumin (OVA; Sigma Chemical, St. Louis, MO) adsorbed to 2mg alum (ImjectAlum; Pierce, Rockford, IL). A booster injection of the same dose of alum-adsorbed OVA was given 7 days later. After etherization dropped 100 ul ovalbumin solution (containing 20μg ovalbumin) into each mouse's nose in successive 7 days. To the dexamethasone intervened group just had been intraperitoneal injection of 0.2ml (3mg/kg) dexamethasone before provocation and the other was the same. The sensitized control animals received the same immunization but inhaled saline instead of OVA-saline. With the flourescent indicator SYBR-green I, real-time PCR assay was applied to test the proliferation CT value of the two gene, objective gene UGRP1 and house-keeping geneβ-actin, at the time 6h and 24h after the last provocation. The value of correction value was calculated by the mean ofΔΔCt. To compare the value of correction value among the sensitized control group,allergic asthma group and DM intervened group. The level of UGRP1 in BALF of each group was assayed by ELISA. All data was expressed with x±s by using SPSS 13 statistical data processing software. The mouse in the same time in different groups was anal by univariate analysis of variance and mouse in the same group at different time line was analyzed by two random samples t test. P<0.05 had the significant difference. Results:(1) Lung pathohistologic analysis: In asthma group, there was hyperemia in the lung and extensive infiltration of inflammation cell around small bronchi and vessel by HE staining.Eosinophilic granulocyte cell appear in alveolar space. The inflammation was less heavy than asthma group in the dexamethasone intervened group and there was no inflammation in sensitized control group.(2)The expression of UGRP1mRNA in the lung:There was no obvious difference on the quantity of the UGRP1mRNA between asthma mouse and DM intervened mouse(P>0.05), both of which were much lower than the control mouse(P<0.05) at 6h; At 24h the quantity of UGRP1 mRNA from asthma mouse was much lower than other two groups. (3) The level of UGRP1 in BALF:At 6h the quantity of UGRP1 in sensitized control group was the highest among all three groups and at 24h the asthma group was the lowest. In DM intervened group the quantity of UGRP1 at 24h was much higher than that at 6h (P<0.05) and the asthma group was the lowest. Conclusions:The expression of UGRP1 in asthma group is lower than the sensitized control group obviously (P<0.05) in any time, DM could promoted the translation of UGRP1 and maintain the stability of UGRP1 and increase its quantity, through which it can inhibit inflammation.
