The methods of isolation and purification such as silicagel column,Sephadex LH-20 and ODS chromatography as well as recrystallization were used and the chemical structures based on physicochemical properties and spectral data were elucidated to isolate and identify the constituents of the fruits of Ligustrum lucidum Ait. On the basis, insulin resistance cell model was established to investigate the effect of compounds from Ligusrtum Lucidum Ait. MTT were used to test the antineoplasmic activity of triterpenes components from Ligusrtum Lucidum Ait. The results showed that eight compounds were isolated and identified.They are oleanolic acid, acetyl oleanolic acid,19a-hydroxy-3-acetyl-ursolic acid, quercetin, apigenin-6"-O-acetyl-7-O-β-D-glucoside,p-hydroxyphenethyl-O-β-D-glycoside,nuezh-enide,GI3. In the non-insulin condition, oleanolic acid,19α-hydroxy-3-acetyl-ursolic acid,apigenin-6"-O-acetyl-7-O-β-D-glucoside,p-hydroxyphenethyl-O-β-D-glycoside,GI3 increased the glucose consumption of insulin resistance HepG2 cells by 11.62%,17.21%,11.85%,12.08%,10.54%, after the insulin added, the above compounds also increased the glucose consumption of insulin resistance HepG2 cells, by 12.95%,13.83%,17.41%,14.26%,10.84%. In the non-insulin condition, Nuezhenide and quercetin did not increase the glucose consumption of insulin resistance HepG2 cells, after the insulin added, they increased the glucose consumption of insulin resistance HepG2 cells by 15.70%,17.04%, and quercetin promoted cell proliferation. MTT showed that the rate of the cell growth suppression were increased in dose-dependent manner. The cell cycle analysis showed that the proportion of cells in G2/M phase were higher than control in a concentration and time dependent manner after treated with 19a-hydroxy-3-acetyl-ursolic acid for 48h and 72h. Oleanolic acid,acetyl oleanolic acid and 19a-hydroxy-3-acetyl-ursolic acid could inhibit the proliferation of Hela and A549 cells. They were determind as the antineoplasmic active components in Ligusrtum Lucidum Ait.
