The Clinic Curative Effect Observation Of Qi Zhu Prescription Treating To PLGC And Experimental Study Of The Anti-tumor Mechanism Of AstragalosideIV And β-elemene

Objective:To observe clinical curative effect on precancerous lesions of gastric cancer treated by Qi Zhu Prescription, and to find the synergistic anti-tumor mechanism of AstragalosideⅣandβ-elemene to human gastric cancer cell line SGC7901 From the perspective of molecular biology.Methods:Clinical part:cases of the patients of precancerous lesions of gastric cancer were randomly allocated to treatment grope (50 cases) treated by Qi Zhu Prescription, and control grope (50 cases) treated by WeiFuChun, and observe the clinic effect after treatment of 3 months. Experiment part:The inhibitory effect of AstragalosideⅣandβ-elemene on viability of SGC7901 cell was assessed by MTTassay.The expression of COX-2 mRNA and protein was determined by RT-PCR and Western blot,respectively.The expression of VEGF and PGE2 in culture medium was detected by ELISA.Results:Clinical part:Clinical research showed that treatment group had symptomatic improvement in gastric abscess, painful abdominal, hypodynamia, anorexia, significantly better than control grope(P<0.05), there was no significant difference in diarrhea and gastric upset, (P>0.05). Comparision of the total integral of symptoms had statistical significance(P<0.05)in Qi Zhu prescription group and control group spectively. the total effective rate in Qi Zhu prescription group was 84%, While the total effective rate of the control group was 60.0%, respectively, the difference had statistical significance(P<0.05). there was significant difference in patho-atrophy,intestinal metaplasia and atypical hyperplasia(P<0.05).Experiment part: AstragalosideⅣandβ-elemene inhibited growth of SGC7901 cell in a dose-dependent manner and down-regulated expression of COX-2,VEGF and PGE2.Conclusion:Qi Zhu Prescription was a effective method in the treatment of precancerous lesions of gastric cancer, with the probably antitumor mechanism was related with inhibition of COX-2 expression,which causes the suppression of its downstream product PGE2 expression and down-regulation of VEGF,therefore decreses tumor growth.
