Objective:This article aims to investigate the possible mechanism and the neuroprotective effect of mailuoning in cerebral ischemic injury.Methods:Kunming mice were used for the experiments (male,126 in total).The mouse middle cerebral artery occlusion (MCAO) model was used. They were randomly divided into three groups, including sham operated group, MCAO models with saline treatment (NS group) and MCAO models with mailuoning treatment (M group).The drug delivery was through tail intravenous,0.015ml/g per day, saline as vehicle, from the beginning of reperfusion to 72h. Neurological deficits, brain water content, infarct volume, membrane potential and oxidative stress productions were measured at 12h,24h and 72h after MCAO, respectively.Results:1) Mailuoning significantly decreased the modified neurological severity score after MCAO at all these time points;2) Mailuoning significantly reduced the brain edema caused by MCAO; 3)Mailuoning also can significantly reduced cerebral infarct, especially at 72h; 4) Mailuoning suppressed the reduction of mitochondrial membrane potential of the neurons in cerebral cortex and internal capsule area after MCAO;5) and besides, Mailuoning significantly down-regulated the contents of 3-NT, HNE and 8-OHdG caused by oxidative stress.Conclusions:Mailuoning protected against ischemic brain injury in mice, improved neurofunction, decreased infarct size, and suppressed edema, and the mechanism might be related to its role in antioxidant.
