Mycelium of Phellinus baumii as research material, the main active ingredient-polysaccharides's extraction, purification, biological activity, physicochemical properties were studied in this paper, and is designed to Phellinus baumii of basic research and application providing a theoretical basis and solid foundation.Using single factor and orthogonal to Phellinus baumii mycelium extraction of polysaccharide as indicator. Test main factors of hot water, ultrasound, ultrasound enzyme three extraction methods affected the extraction yield of mycelium polysaccharide, the results show that the hot water extraction the best process conditions for processing time of 3.5h, solid to liquid ratio 1:50, temperature 90℃, polysaccharides was 2.47%; ultrasonic optimal conditions for the ultrasonic processing time of 20min, solid to liquid ratio of 1:25 power 500W, polysaccharide was 3.356%; ultrasonic compound the optimum conditions for enzymatic pH6.5, hydrolysis temperature of 50℃,2.5% cellulase,2.5% pectinase, protease 1%(% on material), hydrolysis time 120min, was 6.619% polysaccharides.Using trichloroacetic acid (TCA) method, studied the TCA content and times to the removal of protein effect, the results show that when the TCA content of 7.5%, removal of 3 times, get better removal efficiency at this time the removal of protein rate was 83.28%, polysaccharide loss rate was 11.91%.By DEAE-52 anion exchange column and Sephadex G-100 dextran gel column chromatography, anion exchange column and elute by PBⅠ, PBⅡtwo components, dextran gel Sephadex G-100 column chromatography were received PBⅠ-1, PBⅡ-1 two components, identified through a single symmetrical HPLC peak for the homogeneous polysaccharide.Respectively, studied the crude polysaccharide and purification polysaccharides components of immune activity, the results show that crude polysaccharide, PBⅡ-1 mice, significantly increased macrophage phagocytosis of the system, can improve the clearance index and mice phagocytic index, and to some extent in immunosuppressive mice immunity, PBⅠ-1 have some effect but not significant. crude polysaccharide, PBⅡ-1 could significantly promote the hemolysin production, and to some extent immunocompromised mice hemolysin. PBⅠ-1 have some effect but not significant.Physical and chemical properties of different components showed that, PBⅠ-1 molecular weight of about 47,400, while the PBⅡ-1 molecular weight of about 8710. these two components have the characteristic absorption peak of polysaccharides,869cm-1 shows the absorption peak near the polysaccharide isβ-glycosides. PBⅠ-1, PBⅡ-1 by rhamnose, arabinose, xylose, glucose and galactose with a molar ratio, respectively 3.47: 1.99:1:63.27:13.44 and 10.46:1:1.03:82.75:30.94.
