| Objective To establish mouse model of bone marrow transplantation using GFP transgenic mice as donor. To investigate the effect of bone marrow derived stem cells (BMSCs) on the carcinogenesis of mouse gastric cancer mouse induced by 3-methylcholanthrene (3-MCA).Method Six week-old female C57BL/6-GFP mice were used as donor mice, and six week-old male C57BL/6 mice as the recipient for the establishment of bone marrow transplantation model. Peripheral blood was harvested 4 weeks after transplantation and GFP expression cells were assessed by flow cytometries (FCM). Mouse model of gastric precancerous condition was induced using 3-methylcholanthrene (3-MCA). Stomach was collected to detect GFP expression and Y chromosome, presence by FISH.Results1,FCM result indicated that 94.78%±2.5% of bone marrow cells in the recipient were GFP+cell.2,At week 12,80% of mice showed thickening of the gastric mucosa and chronic inflammation. At week 20, atypical hyperplasia was seen in 16.7% of mouse, while the control groups only showed signs of thickening of the gastric mucosa.3,The control group showed only small amount of GFP expressions in interstitial sites between gastric glands, while experimental group there were GFP+cell in the atypical hyperplasia area.4,GFP+ cells were detected in the brain, liver, and kidney within the blood vessels and along the blood vessel walls.Conclusions1,Mouse model of bone marrow transplantation with different gender was successfully established.2,3-MCA-containing silk suture on to the glandular stomach successfully induced gastritis and precancerous lesions.3,BMSCs participated in the process of mucous repair after damage, and cell fusion between BMSCs and gastric epithelial cells occurred during the process of healing, and fused cells gave rise to atypical hyperplasia.4,BMSCs participated in the renewal of blood vessel endothelial cells in different organs.
