| Objective:Osteoprotegerin (OPG) is a member of the tumor necrosis factor receptor (TNFR) family. OPG mRNA expression has been demonstrated in numerous tissues, including endothelial cells and vascular smooth muscle cells. OPG also acts as an important regulatory molecule in the vasculature. it can act as a survival factor for endothelial cells.To investigate OPG expression of aorta endothelialcells and the correlation with other factors in different stage in the diabetes rats. So that it can reflect the diabetes complications of cardiovascular disease at the early time。Methods:forty male SD rats were randomly divided into 4 groups:A,B,C,and D(n=10 each group).Among them A was the normal group, B was insulin resistance group(the diets enriched with fat and sugar),.C was the diabetes for 4 weeks (the diets enriched with fat and sugar for 7 weeks, then low dose STZ induced T2DM rats model, then continue with the diet with fat and sugar for 4 weeks),and D was the diabetes for 8 weeks(the diets enriched with fat and sugar for 7 weeks, then low dose STZ induced T2DM rats model, then continue with the diet with fat and sugar for 8 weeks). At the end of 7 weeks test the trial of OGTT to judge whether they were IR (insulin resistance), and test the insulin and blood glucose, after the low dose STZ induced T2DM rats model, test the plasma glucose,when it was≥16.7mmol/L,the rats were diabetes. Investigate Osteoprotegerin expression of aorta endothelialcells by IH (immunohistochemistry) and test the serum INS (insulin),GSP(gycosyluted serum Portion) and Hs-CRP(high sensitive C reactive protein),and so on.To observe the correlation between these. The data were analyzed by SPSS 13.0 PC program for windows.Resu It:afer 7 weeks,the B group was insulin resistance group.at the end of the trial,The level of OPG expressed in the aorta endothelialcells of the rats were significantly higher in B,C and D group compared with those of A group.(P<0.01),the C and D group were higher the B group((P<0.01);the level of gycosyluted serum Portion(GSP) of C and D group were higher compared with A and B group.(P<0.01),and the D group was higher than the C group(P<0.01).The level of fast insulin(FINS) in B,C and D group were significantly higher than A group(P<0.01, P<0.05, P<0.01),and the B and D group were higher than the C group(P<0.01, P<0.05).The level of FPG in C group was higher than A and B group(p<0.01), D group was higher than A and B group(p<0.01),between the A and B,C and D group, there were no difference. The level of the Hs-CRP in B,C and D group were higher than the A group (P<0.05,P<0.01,P<0.01), the D group was higher than the B and C group(P<0.01);The level of the FFA in B,C and D group were higher than the A group (P<0.05,P<0.01,P<0.01), the D group was higher than the B group(P<0.05);The level of TG in B,C and D group were higher than the A group (P<0.05,P<0.01,P<0.01), the C and D group were higher than the B group(P<0.05); The level of TC in the C and D group were both higher than the A and B group(P<0.01);The level of LDL in D group were higher than the A,B and C group(P<0.01,P<0.05,P<0.05);The level of HDL in A group was higher than the B,C and D group (P<0.05,P<0.01,P<0.01),the B group was higher than the C and D group(P<0.05,P<0.01);The level of VLDL in C and D were both higher than the A and B group(both P<0.01).And OPG was positively correlate with GSP (r=0.579,P<0.01), FINS (r=0.389,P<0.05), FPG (r=0.618,P<0.01), Hs-CRP(r=0.653,P<0.01), FFA (r=0.435,P<0.01), TG(r=0.571,P<0.01) and VLDL (r=0.372,P<0.05);And was negatively correlate with HDL(r=-0.656,P<0.01).Conclusion:(1)Using intragastric administration with fat milk (high-glucose-high-fat diet) plus low dose STZ by vena caudalis, the type 2 diabetes mellitus SD rat model can be induced successfully, the method is reliable, and all the STZ injected SD rats feed with fat milk developed diabetes; (2)The aorta diseases of type 2 diabetes mellitus are advanced with the different course of disease, while OPG acts as an important regulatory molecule in the vasculature, the expression of OPG was different in different stage of the diabetes, and it gradually higher with the stage. (3)Glucose,insulin, lipid metabolism and inflammation contribute to different stage of the diabetes, they can reflect the diabetes vascular complications.(4)And Glucose,insulin, lipid metabolism and inflammation can effect on OPG. And we found that OPG was higher in the stage of prediabetes (insulin resistance), So OPG could reflect the diabetes complications of cardiovascular disease at the early time, it was helpful to diagnosis the complications of the diabetes mellitus. But the retail of the mechanism on OPG expression in the diabetes should be researched in the future. |
