Experimental Study Of Umbilical Cord Blood Mesenchymal Stem Cells As Carrier The Transgenic TNF-α On The Apoptosis Of Nude Mice Gastric Carcinoma Cells

Objective:To research TNF-αclose, sustained, high concentrations of expression in gastric stromal and observe the effect of anti-gastric cancer cells, as MSC as a carrier of TNF-α.Methods:The cultivated positive human gastric cancer cell SGC-7901 was injected into 15 nude mice subcutaneously groin.The model of transplanted human gastric cancer cell SGC-7901 in nude mice was set up.Fifteen tumorigenesis nude mice was separated into three groups randomly:the first group is Mesenchymal Stem Cells from Human Umbilical Cord Blood as carrier of transgenic TNF-α(MSC-TNF-α);the second group is Mesenchymal Stem Cells from Human Umbilical Cord Blood(MSC);the third group is normal sodium(NaCl). Each group has 5 nude mice.The nude mice was observed 14 days.Three groups of nude mice were injected around the tumor with MSC-TNF-α, MSC, and NaCl;the group of MSC-TNF-αand MSC as the treatment group,the group of NaCl as the control group.The transplanted gastric cancer volume and weight were observed in order to find out the difference in the three groups.Executed the nude mice in the 14 day, measured the long and short diameter of the tumor, to obtain the tumor's approximation volumes and the ratio of curbing carcinoma. The expression of TNF-αmRNA in the three groups were determined by fluorescent quantitative PCR method.The expression of TNF-αprotein in the three groups were determined by elisa method. Sliced the tumor tissue into pathological section, observed the necrosis areas in the tumors.Results:The average tumour volumes(cm3)of MSC-TNF-αgroup,MSC group and NaCl group were(1.38±0.21)cm3,(1.80±0.23)cm3 and(2.89±0.29)cm3 respectively.There are differents significantly in the three groups(P<0.001).The treatment with MSC-TNF-αled a strong suppressive effect on the tumor growth compared with MSC and NaCl(P<0.001).Tumour inhibition rate of MSC-TNF-αgroup and MSC group were 52.25% and 37.60% respectively. The results of RT-PCR exhibited that the expression of the TNF-αmRNA were all founded in the three groups.There are differents significantly in the three groups.The results of elisa exhibited that the expression of the TNF-αprotein were all founded in the three groups.There are differents significantly in the three groups. The tumour necrocytosis areas in MSC-TNF-αgroup and MSC group were more than NaCl group.The tumour necrocytosis areas in MSC-TNF-αgroup was more than MSC group.The differents in these groups are significant(P<0.001).Conclusion:The expression of the TNF-αwere all founded in MSC-TNF-αgroup,MSC group and NaCl group.MSC-TNF-αcan obviously kill gastric cancer cells in nude mice.TNF-αand the MSC can cooperate to increase the cytotoxicity of the nude mice gastric cancer cells.
