Objective:To study the effects and mechanism of Shengqi Chinese Herbal Formula on Glucose Consumption in vitro by the serologic pharmacological test.Methods:1. The murine insulinoma cell line INS-1 was individed into 3 groups as normal group, Shengqi Chinese Herbal Formula group and positive control group (Gliclazide). the concentrations of insulin in supernatant were determined by ELISA after stimulated with different concentrations of glucose.2. The international method was used on 3T3-L1 adipocytes differentiating to mature adipocytes and the oil red O staining was used to identify the cell differentiation. After that, We established insulin resistant 3T3-L1 by dexamethasone,the cells were individed into 4 groups as normal group, model group, Shengqi Chinese Herbal Formula and positive control group (rosiglitazone), then we abstract the supernatant to detect glucose Consumption and MTT assay, The mRNA expression of GLUT4 was detected by reverse transcription PCR (RT-PCR).3. The cell line HepG2 was established as insulin resistant model by insulin and was individed into 4 groups as normal group, model group, Shengqi Chinese Herbal Formula group and positive control group (rosiglitazone), then we abstract the supernatant to detect glucose Consumption and MTT assay, The mRNA expression of GLUT2 was detected by reverse transcription PCR (RT-PCR).Result:1. The concentrations of insulin were not significant differences in normal group and Shengqi Chinese Herbal Formula group in different concentrations of glucose. (P> 0.05).Compared with the other groups, the concentrations of insulin in positive control group (Gliclazide)) increased (P<0.05).2. Compared with the model group, the glucose Consumption of 3T3-L1 adipocytes in Shengqi Chinese Herbal Formula group increased (P<0.05). There were no significant differences in Shengqi Chinese Herbal Formula group and the positive control group (P>0.05).The effect of reducing glucose is dependent on concentration, and it declined as glucose concentration increase. Compared with the model group, the mRNA expression of GLUT4 in Shengqi Chinese Herbal Formula group and the positive control group increased (P<0.05).3. Compared with the model group, the glucose Consumption of HepG2 in Shengqi Chinese Herbal Formula group increased (P<0.05). There were no significant differences in Shengqi Chinese Herbal Formula group and the positive control group (P>0.05).The effect of reducing glucose is dependent on concentration, and it declined as glucose concentration increase. Shengqi Chinese Herbal Formula may improve the role of insulin resistance, through the increase of the mRNA expression of GLUT2 and GLUT4.
