Objective:To establish experimental varicocele model in rats to analyze and measure the testis temperature, the the Bcl-2 expression and oxidative stress level to investigate the mechanism of the infertility resulting from varicocele.Methods:The varicocele model was created by partial ligation of the left renal vein in adolescent male Wistar rats. A total of 40 male adolescent Wistar rats were randomly divided into 8-week VC group,12-week VC group and the corresponding sham-operated control group. The rats were killed 8 weeks and 12 weeks after operation, the diameter of the left spermatic vein, bilateral testicular temperature were measured, the Bcl-2 was determined by Immunohistochemistry, the level of H2O2, CAT, SOD and T-AOC were determined by Colorimetry.Results:The experimental varicocele model was successfully created. Compared with the corresponding control group, the diameter of the left spermatic vein enlarged remarkably(P<0.01), bilateral testicular temperature was no significant difference (P> 0.05), The staining indices of Bcl-2 in bilateral testicular mesenchymal cells of the VC groups decreased significantly compared with that in the corresponding control group (P<0.05, P<0.01), and the left sides decreased more significantly than the right sides (P<0.01), the staining indices decreased more significantly in VC12 group than that in VC8 group (P<0.05). The level of H2O2 in bilateral testes in both VC groups increased significantly (P<0.05, P<0.01), and the left sides increased more significantly than the right sides (P<0.05, P<0.01), increased more significantly in 12-week VC group than that in 8-week VC group (P< 0.05); The activity of CAT reduced significantly in left testes (P<0.05), the right sides had no statistical significance (P>0.05), but the numeric had already a downward trend; The SOD activity and T-AOC of the bilateral testes in both VC groups reduced significantly compared with that in the corresponding control group(P<0.01), and them reduced more significantly in 12-week VC group than that in 8-week VC group (P<0.01), the T-AOC of the left sides were lower than the right sides in both VC group (P<0.01).Conclusion:We made a preliminary observation and research on experimental varicocele model, considered that the VC can't lead to bilateral testicular temperature change remarkably, but the VC caused bilateral testicular H2O2 content increased, Bcl-2 expression, CAT activity, SOD activity and T-AOC declined, made cell apoptosis decreased, the oxidative stress system disturbance, and made testicular damage significant increase as the duration of varicocele gradually increased.
