Biology And The Sclera Morphology Changes In TIMP-2 Gene-transferred Guinea Pig Form Deprivation Myopia Model

Abstract/Summary:
Objective To observe the axial length, refractive error and the morphology changes of TIMP-2 gene-transferred guinea pig form deprivation myopia model. Explore if the TIMP-2 gene can affect to the guinea pig form deprivation myopia model.Methods 60 four weeks of age guinea pigs were randomly divided into four groups(15). Right eyes of the experimental groups were covered a translucent plastic occluder, the left eyes were opened. Gene-transferred liposome, empty plasmid- transferred liposome and normal saline will be injecteded in the first three groups respectively on the fifith day. Refraction,axial lenth,structure and ultrastructure of sclera were measured respectively after the injection 2,7,14 days. The sclera will be examined with light and transmission electron microscope.Results The axial length and degree of myopia changed rapidly in the first week,then slowing down and stabilizing. On the 2,7,14 day after injection, the average degree of myopia of the TIMP-2 group are -0.4±0.5D,0.9±0.6D,1.2±0.6D. Compared with the other three groups, the differences of the14 days are statistically significant(P<0.05). On the 2, 7,14 day after injection, the axial length of the experimental group are 7.78±0.04mm, 7.69±0.03mm,7.65±0.03mm. Compared with the control group, the differences of the 7,14days are statistically significant(P<0.05). Histopathologic examination showed thicken of posterior sclera after injection. Collagen fibrillar diameters of sclera in the posterior segment of formed deprivation were obviously bigger and the space between the fibers was smaller than that of control eyes. TIMP-2 group of the right deprivation eyes average diameter fibers are 49.7nm,71.3nm,97.8 nm respectively. Compared with the control group, the differences of the 7,14 days are statistically significant(P<0.05).Conclusion TIMP-2 gene transferres guinea pig form deprivation myopia model.The sclera shows thickening. The axial length become shorter. Collagen fibrillar diameters of sclera become bigger. The degree of myopia go down. The changes of the two sides have the same inhibitory effect to the myopia. The sclera morphology changes faster than the biology. Gene TIMP-2 has inhibitory effect to the changes of biology and morphology in guinea pig form deprivation myopia model.
