Tagert: ADSCs will be separated from the rat's adipose, cultivated and observed in vitro. At the same time, ADSCs will be induced to neurocyte differentiation and the signs of early nerve-cell reflected by ADSCs after the induce will be measured. Afterwards, the influence on the rat with craniocerebral injury after the ADSCs transplantation and the neurocyte apoptosis as well as survivals of the transplanted cells will be observed. Significance: Full discussion on the feasibility of adopting ADSCs transplantation as a new method for treating craniocerebral injury Methodology: The model of the rat with craniocerebral injury will be made through free falling body method. 60 healthy female wistar rats will be divided by digital method at random into transplantation experimental group, cultivation experimental group and sham operation group, each of which has 20 rats. Brdu marks the cells transplantation of injury areas of the rats from the transplantation experimental group by ADSCs; the cultivation experimental group transplants DMEM/F12 culture medium of equal quantity; and the sham operation group only executes craniotomy without transplantation. After the transplantation with NSS, 1,3,10,21d respectively scores the rats of each group. Brain tissue of the injured area will be taken and the survival and proliferation of the cells in the area will be examined and marked through direct immunofluorescence. TUNEL will be employed to observe the cell apoptosis.Result: After transplantation, ADSCs, distributed in the injury area, survive well. In 3, 10, 21 d NSS, the score of the transplantation experimental group is less than that of the cultivation experimental group (P < 0. 05). In each time interval, the cell apoptosis of the transplantation experimental group is evidently less than that of the cultivation experimental group (P <0.05).Conclusion:After ADSCs transplantation, the cell apoptosis of the rat with craniocerebral injury will be decrease, and it is helpful to the recovery of neurological function.
