| Objective: To evaluate the reliability of the model of posterior circulation ischemia in rabbits made by deligation in three blood vessels. Methods: The 108 New Zealand rabbits were divided into 15 minutes (n=36), 30 minutes (n=36) and 120 minutes (n=36) after operation. The rabbits of above-mentioned time were matched according to sex, age, and weight, then were randomly divided into the group of sham operation (n=12), the group of deligation in right common carotid artery (n=12) and the group of deligation in three blood vessels (right common carotid artery and double vertebral arteries) (n=12). The physiological indexes(heart rate, respiration rate, pupil diameter) of every group were examined and these rabbits were examined with PWI and DWI according to homologous time. After examination two rabbits'brains of every group were rapidly taken through decapitation and sliced according to the deck of MRI. The brain slices were stained in the liquor of two percent TTC. Another four rabbits'hearts were perfused with four percent paraformaldehyde and the ROI of two brains were taken to prepare for the specimens of pathology and the other for the specimens of electronmicroscope. Results: The physiological indexes of every group in homologous time had no significant difference (P>0.05). At 15 minutes after operation, the CBF decreased, the MTT extended in the rabbits of the group of deligation in three blood vessels and had significant difference (P<0.001) versus other two groups respectively, but the CBV and ADC had no significant difference (P>0.05). At the 30 minutes after operation, the CBF, the CBV decreased, the MTT extended in the rabbits of the group of deligation in three blood vessels versus other two groups respectively and had significant difference (P<0.01), but the ADC had no significant difference (P>0.05). At the 120 minutes after operation, the CBF, the CBV decreased, the MTT extended, the ADC decreased in the rabbits of the group of deligation in three blood vessels and had significant difference (P<0.01) versus other two groups respectively. The CBF, CBV, MTT, ADC in the group of deligation in right common carotid artery had no significant difference (P>0.05) versus the group of sham operation. The DWI had no abnormal high semaphore in every group at above-mentioned time. The electronmicroscope examination was abnormal in the group of deligation in three blood vessels, but nerve cells had no necrotic structural alteration. TTC staining and the pathologic examination (HE dyeing) were normal in every group. Conclusion: The model of posterior circulation ischemia in rabbits can be made by the deligation in three blood vessels, but infarction isn't found in the model. |
