Objective:To investigate the clinical effect of the therapy of arimedex (Letrozole LE) in induction of ovulation in polycystic ovary syndrome(PCOS).Methods: 189 consecutive infertile patients with PCOS were recruited. 68 patients were randomized to the LE group (178 cycles) and the 96 patients were recruited to the Chloramiphene (CC) group (189 cycles). Main outcome measure(s): growth rate of follicles, serum hormone level, endometrial thickness, cervix mucus, ovulation and pregnancy rates. Results: The growth rate of follicles in LE group and CC group were 1.67mm/d and1.78mm/d (P=0.14). The level of serum T on the day of LH peak in LE group and CC group were 2.1±1.9 ng/dl,2.0±1.7ng/dl (P=0.93). The LE group's serum T level in on the day of LH peak and on day 3 were 2.1±1.9 ng/dl,1.4±1.5 ng/dl (P=0.09). The level of serum E2 on the day of LH peak in LE group and CC group were 89.0±59.5pg/ml, 108.1±46.0pg/ml (P=0.13). The endometrial thickness in LE group and CC group were11.3±1.6mm,9.7±1.4 mm (P=0.07) .The cervix mucus in LE group and CC group were11.0±1.1,8.3±3.1 (P=0.003) . Conclusions: Letrozole was benefit for ovulation induction in patients with polycystic ovary syndrome, and it didn't aggravate the level of serum T abnormality.
