Objective:To investigate the role of cytokine inducer polyinosinic on duck hepatitis B virus and the mechanism.Methods: The random primer method was selected and the DHBV DNA was marked with digoxigenin and Chongqing duck serums were detected by dot blotting. The positive serums were selected to artificial infected the one day old Chongqing duck in order to establish animal models. Then,the animals were randomly divided into phosphate buffered saline (PBS) control group and Polyinosinic treatmental group with intraperitoneal injection 20μg/Kg once a day for consecutive three days.At the same time set the hidden ducks uninfected control group. The blood was drawed off from the external jugular vein on before treament,medication three days and the serum was separated and stored at -20℃for inspection. The serum TNF-α,INF-γand The O.D values DHBsAg in duck serum and The serum AST,ALT were observated.The liver tissue was extracted on before treament,medication three days.The paraffin section was prepared to observed the pathological changes of liver tissue.The immunohistochemical was used to observed the change of the number of antigen-positive cells in the liver cell.Results: Serological observations:One,Duck liver function:There is no significant improvement in liver function after three days treament of polyinosinic.Two, DHBsAg level: The O.D values DHBsAg in duck serum has no change.Three, Cytokine level: Polyinosinic could increase significantly serum TNF-αand INF-γ. Histopathological observations:The necrosis of duck liver has no change after three days treament of polyinosinic. Immunohistochemical observations: The number of S antigen-positive liver cells has no reduce after three days treament of polyinosinic.Conclusion: Dose Polyinosinic of short course of treament can induce serum TNF-αand INF-γincrease significantly,but no inhibition on duck hepatitis B virus.
