| Objective: In order to further clarify the issues on which DangGuiNianTongTang of rheumatoid arthritis-the anti-inflammatory and analgesic effects,using AA rats by different doses of DangGuiNianTongTang after treatment, by detecting matrix metalloproteinase-3, C-reactive protein, erythrocyte sedimentation rate , joint pain index, weight and other indicators of the changes, designed to explore the progression of early control of the effective herbs, and a better understanding of different doses of DangGuiNianTongTang of rheumatoid arthritis-related indicators and the improvement in symptoms. Methods: (1) Select Wistar rats 60, male, according to random number table, divided into blank group, model group, DangGuiNianTongTang low dose group, DangGuiNianTongTang mid-dose group, DangGuiNianTongTang high-dose group , each 12, and number, in addition to the blank group, the rats were each injected into the right rear toe skin 0.1m Freund's complete adjuvant-induced inflammation. (2) Two weeks after making model, all rats were taken by gavage, a total dose 18 days, blank group and model group were given distilled water and other capacity. (3) Modeling and medication before and after the observation of the general situation in rats, recording weight, caused by inflammation at different times after the ankle joint swelling, joint pain index, with Clostridium Measuring ESR, measured using ELISA, matrix metalloproteinase-3, C-reactive protein, and the right ankle to get fixed, decalcified, embedded in paraffin, HE staining, light microscopy observation of pathological changes in joints. (4) Application SPSS13.0 statistical software for statistical analysis. Results: (1) Of matrix metalloproteinase-3, C-reactive protein, erythrocyte sedimentation rate in the dose group and high-dose group was not statistically significant (P> 0.05), medium-dose group, high-dose group and low-dose group were statistically significant (P <0.05). (2) After inflammation, at different times the changes of swelling ankle and joint pain index have interaction intreatment methods and time;There are no different between high and middle dose group(P>0.05) ;but compared to model and low dose group,in high and middle dose group,the difference exist(P<0.05). (3) After inflammation, at different times the changes of body weight, there are signifficent difference when the middle group,compared to high and low dose,model group,the difference exist (P<0.01). (4) DangGuiNianTongTang inhibit certain destructive synovium, in which DangGuiNianTongTang, high-dose and mid-dose group was significantly lower doses. Conclusions: (1) DangGuiNianTongTang mid-dose group has a good anti-inflammatory analgesic effect, and the swelling with high-dose group was no significant difference in pain. (2) DangGuiNianTongTang inhibit certain destructive synovium, in which DangGuiNianTongTang, high-dose group was significantly lower doses. (3) DangGuiNianTongTang dose group gastrointestinal damage small, medium-dose group body weight than the other model rats significantly increased body weight, indicating that the party has a good dose of original work to reconcile the role of the spleen and stomach, spleen and stomach medicine is the source of qi and blood biochemistry, there are indeed practical significance. |
