Objective To study the relationship between the mortality of cells and the expression of PUMA gene on p53 wild type LNCaP cells which were induced by different doses of 60Coγ-ray. To research the effect of PUMA gene on human prostate cancer cells.Methods Using MTT assay to detect the proliferation of the LNCaP cells which were cultivated 0h,24h,48h,72h induced by different doses of 60Coγ-ray; Using MTT assay to detecte the mortality of the human prostate cancer LNCaP cells which were induced by different doses of 60Coγ-rays in vitro; Using reverse transcription -polymerase chain reaction(RT-PCR)assay to detecte the expression of PUMA gene on LNCaP and PC-3 cells induced by 0Gy,3Gy,5Gy,8Gy doses of 60Coγ-rays and to study the relationship with the mortality of cells.RESULTS The proliferation of the test group's LNCaP cel(l3Gy,5Gy,8Gy)is lower than the control group(0 Gy)cell's in the same doses of 60Coγ-rays irradiation and different incubation time (0h,24h,48h,72h )after exposed by 0 Gy,3Gy,5Gy,8Gy doses 60Coγray(P<0.01); The mortality of the test group LNCaP cell is higher than the control group cell's in the same incubation time and different doses of 60Coγ-rays irradiation(P<0.01),with the irradiation of 60Coγ-rays does higher,the mortality of LNCaP cell become more high (P <0.01); With the irradiation of 60Coγ-rays does higher,the PUMA-mRNA of LNCaP expression become more obvious(P <0.05), the control group and test group could not see the PUMA-mRNA on the PC-3 cells.Conclusions The expression of PUMA,induced by 60Coγ-rays,plays an important role in mediating LNCaP cell death.
