Objective To study the effect of propofol general anesthesia on the learning and memory function of the first trimester pregnancy Sprague-Dawley rat's offspring.Method After confirmation of successful mating,20 femal SD rats were randomly divided into propofol group (group P) and control group (group C) .Propofol was infusded at a rate of 20 mg/(kg·h) for 2 hours after the induction dose of 20mg/kg via tail vein when they were in the 7 th day of their gestation in group P. While in groupC, propofol was replaced with eaqual volume physiological saline.The learning and memory function of the offspring rats were tested by Morris water maze test, and the hippocampus tissue was collected for HE staining and electrical microscope at postnatal time 30 days,60 days and 90 days respectively. The expression of the immediate early genes ,c-fos and c-jun mRNA were measured by RT-PCR.Result The seeking-platform latency,active time in the second quadrant and number of crossing-platfom had no singnificant difference between group P and C at the observation time (P >0.05). HE staining and electrical microscope of hippocampus tissues shows significant difference between two groups. The expression of the c-fos,c-jun mRNA were appeared in the both the offspring of the two groups and their gay shows significant difference between two groups(P >0.05).Conclusion Propofol has no effect on learning and memory function of the first trimester pregnant Sprague Dawley rat's offspring in this study.
