Objective:To evaluate the protective effect of trimetazidine combined with benazeprilon soproterenol-induced myocardial injury of rats.Methods:Forty rats were randomly divided into five groups as follow:NS group, ISO group, TMZ group, Ben group and TMZ+Ben group. There were eight rats in each group. Rat cardiac injury models were estabished by subcutaneous injection of isoproterenol. In the experiment, NS group were intragastric administrated with saline,from the fouth day to the tenth day, the rats were injected with saline after they were intragastric administrated 30 minutes. ISO group, TMZgroup, Ben group and TMZ+Ben group were intragastric administrated with saline,trimetazidine 10mg/(kg·d),benazepril 10mg/(kg·d),trimetazidine 10mg/(kg·d) and benazepril 10mg/(kg·d) respectively from the first day to the third day, from the forth day to the sixth day,all the rats were injected with Isoproterenol 20,10,5 mg/(kg·d), from the seventh day to the tenth day, all the rats were injected with Isoproterenol 3mg/(kg·d).After 6 weeks, the ratio of heart weight to body weight (HW/BW)and left ventricular weight to body weight (LVW/BW) were caculated,the pathological changes of myocardial tissue were observed, myocardial apoptotic index (MAI) was detected by TUNEL.Results:Compared with the ISO group, trimetazidine and benazepril could markedly reduce myocardial necrosis,reduce interstitial edema. HW/BW and LVW/ BW in ISO group, TMZ group, Ben group and TMZ+Ben group were statistically significant compared with NS group (P<0.05),those in TMZ group, Ben group and TMZ+Ben group were statistically significant compared with ISO group (P<0.01), those in TMZ+Ben group were statistically significant compared with TMZ group and Ben group (P<0.05),and those in TMZ group were no statistically significant compared with Ben group (P>0.05), The MAI of ISO group,TMZ group,Ben group and TMZ+Ben group was significantly higher than NS group(P<0.01), that of TMZ +Ben group was significantly lower than those of ISO group,TMZ group,Ben group (P<0.01), that of TMZ group was no significantly different compared with that of Ben group (P>0.05).Conclusion:Trimetazidine and benazepril can significantly reduce the isoproterenol-induced myocardial necrosis,myocardial hypertrophy, reduce myocardial apoptosis, and their combination can protect myocardium betterly.
