Objective:To detect the expression of PRMT2 and its variants in breast carcinoma tissue and adjacent tissue, then discussed the relationship between the proteins and other clinical parameters.Methods:1. The expression of PRMT2 and its variants in breast cancer and pairing adjacent tissue was detected by Western blot; 2. The expression and subcellular localization of PRMT2 and its variants PRMT2βin breast cancer cell lines (MDA–MB -231, T47D)were detected by immunofluorescence; 3. Collected 63 cases of breast malignant tissue and 20 cases of adjacent tissue, detected the differences of the expressing levels of PRMT2 and its variants PRMT2βbetween malignant tissue and adjacent tissue by immunohistochemistry, then discussed the relationship between the proteins and clinical parameters in breast cancer.Results:1. Western blot showed that:PRMT2 and its variants were expressed in breast cancer and pairing adjacent tissue, the expressing levels in breast cancer was much higher than in the pairing adjacent tissue.2. Immunofluorescence showed that:In breast cancer cell lines, PRMT2 protein distribution within the cytoplasm and main in the nucleus; while PRMT2βprotein scatter homogeneously in nuclei and cytoplasm.3. Immunohistochemistry showed that:In carcinoma cell and adjacent cell, positive signals of protein PRMT2 and the variants PRMT2βwere brown granules, which located in cytoplasm, nuclei excepted the nucleolus had little diffusion to scatter. In 63 cases of breast malignant tissue, the positive rate of PRMT2 expressing was 96.8%, and in 20 cases of adjacent tissue, the positive rate of PRMT2 expressing was 50.0%; the positive rate of PRMT2βexpressing was 93.6% in breast malignant tissue,and, PRMT2β's positive rate in adjacent tissue was 45.0%. There were significant differences of PRMT2 and PRMT2βexpressing levels between malignant tissue and adjacent tissue(P<0.01).4. Immunohistochemistry showed that:In 63 cases of breast malignant tissue, the expressings of PRMT2 and its variants PRMT2βshowed no relationship with patients'age, size of tumour, menopause, histologic typing, metastatic lymph node, TNM clinical stage, ER, HER2, Pgp (P>0.05). The expressings of PRMT2 showed no relationship with PR, The strong positive rate of PRMT2βin PR(-)of carcinoma was 30.0%; while in PR(+)of carcinoma was 65.1%, which showed significant difference(P=0.009).Conclusion:1. The subcellular localization of PRMT2 and its variants PRMT2βexisted difference between the human breast cancer cell lines and cancer tissue cells.2. PRMT2 and its variants PRMT2βexpressing levels in breast malignant tissue were much higher than in adjacent tissue.3. Expression of PRMT2 and its variants PRMT2βhad no significant correlation with positive ER status in breast cancer.4. PRMT2βexpressing levels in PR(+) breast malignant tissue were much higher than in PR(-) breast malignant tissue .
