AIM:To investigate the effects of Ginsenoside-Rg3 with different concentrations in vitro conditions of high glucose in human retinal vascular endothelial cells (HRCECs) proliferation and intercellular adhesion molecule-1 (ICAM-1) expression impact.METHOD:HRCECs from eyeball after cultured in vitro,which being at the 3-4rd gengeration and good growth were taken in experiment.The experiment subjects were divided into low glouse control group,high glucose control group,and high+different concentration Rg3 group, The effect of Rg3 on the proliferation of HRCECs was measured by MTT assay. observe the division HRCECs ICAM-1 expression by Immunocytochemical method.RESULT: MTT colorimetric assay results showed that: the control group with high glucose sugar control group no significant differences (P >0.05), With 10,20,40,80,160mg/L to deal with the ginsenoside Rg3 under high glucose HRCECs, after 24, 48, 72 h, high glucose + different concentrations of Rg3 group with high glucose compared to the control group significant difference (P <0.05), high glucose+ Different concentrations of Rg3 comparison between time and concentration groups are also significant differences(P <0.05). The inhibition rate were 10.25%,18.57%,27.34%,35.22 %,46.66% after they were exposed with Rg3 for 24 hours, they are 26.43%,35.59%,43.82%,50.60%,60.03%for 48 hours and 42.23%,54.47%,64.33%,71.33%,77.87% for 72 hours.Immunocytochemical detection showed that: compared with the control group of low-sugar, high glucose control group, ICAM-1 expression was significantly (P <0.05), With 40,80,160 mg/L to deal with the ginsenoside Rg3 under high glucose HRCECs48h,different concentrations of high glucose + Rg3 group and high glucose control group compared to the expression of ICAM-1 were significantly different (P <0.05), different concentrations of high glucose + Rg3 comparison between groups are significantly different (P < 0.05).CONCLUSION:Ginsenoside Rg3 inhibited under high glucose retinal vascular endothelial cell proliferation and ICAM-1 expression.
