It is clear that c-Myc plays an important role in mice embryonic development. As an appendant mini organ of skin, hair follicle morphogenesis is initiated in the intermediate of embryonic period. However, if c-Myc participates in this development process and what role it plays are still unknown. Previous studies had showed that there was c-Myc expression in adult hair follicle during hair cycle, but its precise position is still controversial, and data in every stage of hair cycle is insufficient. What is more, c-Myc expression pattern in mice and its founction to the growth of hair regenerative process are not clear.In order to explore if c-Myc is involved in mice hair follicle morphogenesis and regeneration in mice natural and synchronized hair cycle, and its role in this process, we detect c-Myc expression pattern with IHC, mRNA and protein level with RT-PCR and western blot respectively. In vivo study, we use specific inhibition polypeptide to c-Myc to interfere normal c-Myc protein expression; on the other hand, we use expression vector to make c-Myc overexpression topically, then observe the effect on hair follicle and check correlation factors. The main results are as follows:1. c-Myc continually expresses in every hair cycle stage of mice hair follicles during its morphogenesis.2. We confirmed the expression pattern of c-Myc in mice natural and synchronized hair cycle. The expression of c-Myc is specifically located in the matrix and inner root sheath (IRS) in anagen, rudimentary IRS in catagen and not detected in telogen.3. Through the specific inhibition to c-Myc protein function topically, we found that the treated hair follicle grew slower and became smaller in size.4. Through the overexpression of c-Myc topically in synchronized hair follicle, we found that the treated hairs grew faster. However, it cannot restart a new hair cycle. In summary, our study confirmed the c-Myc expression pattern in hair follicle morphogenesis and regeneration. The c-Myc expression in certain hair cycle stage and structure exhibit its important role. Inhibition of c-Myc protein result in hair follicle growing slower and becoming smaller, expression of correlation factors became changed. Overexpression of c-Myc lead to accelerated hair growth illustrating that c-Myc plays an important role in hair follicle morphogenesis and regeneration.
