The traditional Chinese medicine, Hui huisuanzi, is the dry and ripe seed of the plant Rannunculus chinensis Bunge. It has been used for anti-inflammatory and alexipharmic drug in many local traditional medicinal systems, such as Chinese herbal medicine.Modern pharmaceutical studies indicate that the extract of Rannunculus chinensis Bunge has protective effect for liver damnification, and preparation of Rannunculus chinensis Bunge has been used in clinic a little. Hoever, the base-substances for anti-HBV has not been elucidated yet. In order to find constituents against HBV of Rannunculus chinensis Bunge, our dissertation choose HepG2.2.15 cell as model and secretive changes of Rannunculus chinensis Bunge as astimate guide-line, aim to find effective constituents from Hui huisuanzi. The dry and ripe seed of Rannunculus chinensis Bunge, is extracted by 90% EtOH, then re-extracted with ethyl acetate, n-butanol successively to give three fractions, among of which, fraction ethyl acetate was the most effective one.The compounds were separated and determind from fraction ethyl acetate, including (1)luteolinluteolin, (2)kaempferol, (3)quercetin, (4)quercitrin, (5) kaempferol-3-O-p-rutinoside, (6) tamarixetin-3-rutinoside, (9) p-hydroxybenzcic acid, (10) methyl 4-hydroxybenzoate, (11) gallic acid, (12) protocatechuic acid, (13) ellagic acid, (14) 3,3'-di-O-methylellagic acid, (15) caffeic acid, (16) ursolic acid, (17)β-ursolic acid, (18) stigmasterol, (19)β-daucossterol, (20) emodin, (21) glucoemodin, (22) 7-ketologanin。Compound (1)~(16), (20)~(22) was found from Rannunculus chinensis Bunge for the first time, Compound (22) was found from genus Rannunculus for the first time.The compounds separated above were screened by HepG2.2.15 cell model, seven of which showed remarkable anti-HBV activities. Compound (4),(3),(5) showed strong inhibition for the secretion of HBsAg, IC50 were 78.4,23.land 52.6μg/ml respectively. Compound(11),(12),(15) showed inhibition in some extent for the secretion of HBeAg, IC50 were 24.5,34.2 and 24.1μg/ml respectively. The anti-HBV activities of compounds were indicated firstly.
