Angina pectoris is a disease of caused by coronary insufficiency, cardiac sharp, temporary ischemia and hypoxia and its main clinical manifestations are syndrome chest pain or chest discomfort. New Guanxinsuhehuoxuefang(NGXHXF) usually used to cure acute AP attack following the theory of traditional chinese medicine,contains Styrax, Borneolum, Olibanum, Radix aristolochiae, Lignum santalialbi, having the power of regulating vital energy , eliminating stasis to active blood circulation and acesodyne.The studys on pharmacodynamics of the NGXHXF has been done. The experimental studies was to use the Pit or ISO copy the mouse and rat experimental acute cardiac ischemia modle. the results showed that the parties to the pit, or iso in the laboratory rats mouse but there's a shortage of the myocardium model better protection, the mechanism may inhibit freedom and the it and protect and improve nappi, diastolic coronary artery etc. At the same time, the experimental results show that the new Guanxinsuhehuoxuefang could reduce the ECG change of j piont and p wave by ISO. there's a shortage of the myocardium, be good resistance to the myocardium, could lower the canister GOT activity (p < 0.05, p < 0.01), Alleviate the cell membrane, the protection of the cell membrane structure and cardiac muscle cells and the myocardium was poorly enzymes the leak to the myocardium was poorly ; SOD activity has raised, the MDA level to reduce significantly reduced, it is to prevent damage, and necrosis myocardium cell, there's a shortage of damage to the myocardium. The NGXHXF content of serum NO significant increase, to effectively protect and improve nappi functions, promoting no generation and released, relaxation of the vessels and their resistance to the myocardium was poorly there's a shortage. The NGXHXF could reduce ISO there's a shortage of the myocardium was poorly model rats content of serum GOT to the myocardium in the organization to spare NO, NO legal activity, the MDA level, the experimental acute myocardium there's a shortage of protection.In view of preminilary study, in this study, we observe mainly the NGXHXF's effect on the isolated heart and blood vessels inotropism of rat,mouse and rabbit. The purpose of choosing isolated heart to study is to find the direct effect of drugs on the heart, especially on the direct impacts on systolic performance.The copy method of isolated heart specimens is very mature and need a simple sample preparation and can last a long time,with good stability,never influenced by HR, coronary vasomotor and nerve humoral regulation and other factors; Measurement and analysis the change of myocardial contractility are more easier than complete hearts, suitable for observation of direct effects of drugs on the heart。Myocardial contractility can be influenced not only by cardiac load ,but also by activity and structure of their own , such as the Ca concentration, glucose, fatty acid metabolism.Isolated heart experiments can evaluate the determinants of cardiac function -- preload, afterload, myocardial contractility, HR, under the conditions of other factors be fixed. Vitro experiments of blood vessels can be more intuitive in observation of vascular reactivity, on other factors fixed conditions,so we used isolated animal heart and blood vessels to do the experiment in this study.PartⅠ: the new Guanxinsuhehuoxuefang(NGXHXF)'s effect on the isolated heart inotropicObjective:observe the NGXHXF's effect on the isolated heart inotropic of rats,mouses and rabbits.Methods:Chose 20 Wistar rats,20 mouses and 20 rabbits, the animals were fed with nutrient food taking food and water freely, and they were raised in sanitary experiment lab with temperatre between 22℃to 26℃.After a week,used 10% urethane (1g/kg) intraperitoneal injection to anesthetize the rats,opened the thoracic cavity immediately, cut the blood vessels connect to heart,took out the hearts of rats,mouses and rabbits,put its into low-temperature oxygenated Krebs liquid,pressed the heart slightly to let the blood out,cut the left and right atrials,fixed one end with thread to the bottom of Magnus bath containing Krebs liquid saturated with 20ml by 95% O2 +5% CO2 mixed gas, connected the other end to tension transducer,used the super constant temperature trough with circulating water to maintain at about 34.5 ~ 35.5℃, adjusted the pre-load until myocardial contractile force reached the maximum,balanced for 15 minute. Used the 10-8mol / L of calcium to live specimen for three times,give the NGXHXF 0.02ml (finalconcentration100mg/l),after 15 minutes used Krebs liquid to wash 3 times, balanced for 15 minute, like this give the new Guanxinsuhehuoxuefang150mg/l(equivalenttodrugs0.03ml),200mg/l(equivalent to drugs0.04ml),250mg/l(equivalent to drugs0.05ml),using MS4000U-1C biological signal quantitative recording and analyzing system to collect the data of the different doses of the NGXHXF changed the atrial muscle tension,and analyzed its effects.Results: (1)The NGXHXF has lower effect on the rats myocardial contractility and HR with the increase the dose of drugs.and has no effect wih 100mg/L(P>0.05); With the increase of dose such as 150mg/L, 200mg/L, 250mg/L,it decresed myocardial contractility and slowed down HR obviously(p<0.05 or p<0.01).(2) The NGXHXF has little effect on the mouses myocardial contractility and HR as the increase the dose of drugs.and has no effect wih 100mg/L(P>0.05); With the increase of dose such as 150mg/L, 200mg/L, 250mg/L,it decresed myocardial contractility and slowed down HR obviously(p<0.05 or p<0.01).(3) The NGXHXF has little effect on the rabbits myocardial contractility and HR as the increase the dose of drugs.and has no effect wih 100mg/L(P>0.05); With the increase the dose of 150mg/L, 200mg/L, 250mg/L,it decresed myocardial contractility and slowed down HR obviously(p<0.05 or p<0.01).Conclusions: (1)The NGXHXF has the effect of decrese on rats,mouses,rabbits myocardial contractility and slow down HR,threrfor improve or eliminate symptoms of angina pectoris. (2)With the increase the dose of 150mg/L, 200mg/L, 250mg/L, ,its effect of decrese myocardial contractility and slow down HR increased obviously.PartⅡ: the new Guanxinsuhehuoxuefang(NGXHXF)'s effect on the isolated blood vessels inotropicObjective:observe the NGXHXF's effect on the isolated blood vessels inotropicMethods:Chose 20 Wistar rats,equal of male and female,weight from 180g to 220g.Rats were fed with nutrient food taking food and water freely, and they were raised in sanitary experiment lab with temperatre between 22℃to 26℃.After a week,used 10% urethane (1g/kg) intraperitoneal injection to anesthetize the rats,opened the thoracic cavity immediately, cut thoracic aorta and abdominal aorta, put them into low-temperature oxygenated Krebs liquid, tried to cut the connective tissue around the artery, cut into 0.7 ~ 1.4cm long,used two triangles made of stainless steel filaments pass through the blood vessels , fixed one end with thread to the bottom of Magnus bath containing Krebs liquid saturated with 20ml by 95% O2 +5% CO2 mixed gas, connected the other end to tension transducer,used the super constant temperature trough with circulating water to maintain at about 34.5 ~ 35.5℃, adjusted the pre-load until myocardial contractile force reached the maximum,balanced for 15 minute. Used the 10-8mol / L of Iso to live specimen for three times , give the NGXHXF 0.02 ml (finalconcentration100mg/l),after 15 minutes used Krebs liquid to wash 3 times, balanced for 15 minute, like this give the new Guanxinsuhehuoxuefang,usingMS4000U-1C biological signal quantitative recording and analyzing system to collect the data of the different doses of the NGXHXF changed the vascular contractility,and analyzed its effects.Results: NGXHXF has higher effect oncontractility of thoracic aorta and abdominal aorta with the increase of dose. Not obviously effest with the dose of 100mg/L (P>0.05);with the dose of 100mg/L, 150mg/L,250mg/L has the obviously effests(p<0.05 or p<0.01).Conclusions: (1)NGXHXF has the effect of decrese the main arteries contraction force of thoracic aorta and abdominal aorta on rats,and the market is playing an increasingly fundamental role in the main arteries contraction force of thoracic aorta and abdominal aorta with the dose of 100mg/L, 150mg/L,250mg/L.(2) The NGXHXF could reduce the main arteries resistance, reduce the load and the myocardium and oxygen, to dissolve angina.(3) The effect of NGXHXF on the main arteries contraction force of thoracic aorta and abdominal aorta contractility correlated with dose.
