Role Of Radix Pulsatillae Alcohol Extract Protecting Tight Junction Protein Between The Epithelial Cells Of Intestinal Mucosal Barrier In Rats With Trinitro-benzen-sulfonic Acid Induced Colitis

Objective:To investigate the effect of radix pulsatillae alcohol extract regularing the tight junction protein of intestinal mucosal in rats with trinitro-benzen-sulfonic acid(TNBS) induced colitis. To observe the expression of the tight junction portein, so as to interpret further the function of radix pulsatillae clcohol extract protecting the intestinal mucosal barrier. Methods:Acute colitis was induced by giving TNBS. Thirty-eight rats including male and female were randomized into normal group(n=8), TNBS group (n=10), radix pulstillae alcohol extract treated group(n=10) and live combined bifidobacterium and lactobacillus tables(Golden Bifid) treated group(n=10). Diarrhea and bloody stool as well as colonic histology were observed. The levels of tumor necrosis factor-a(TNF-a) and interleukin-10(IL-10) in colonic tissue and endotoxin in blood were determined with ELISA. The expression of Occludin between the epithelial cells of intestinal mucosal barrier was detected by histochemical stain method. Results:After acute colitis was induced by giving TNBS, the levels of the inflammatory symptoms and histological damages of colonic mucosa were increased in radix pulstillae alcohol extract treated group and golden bifid treated group, while they were obviously decreased (9.20±1.75 Vs 6.50±1.27,5.90±1.67, P< 0.05; 7.10±0.99 Vs 5.00±1.05,4.80±1.23, P< 0.05). Compared with TNBS group, the levels of TNF-a and endotoxin in blood were significantly reduced (657.54±149.60 Vs 521.24±109.38,514.73±163.47, P<0.05; 0.213±0.023 Vs 0.148±0.093,0.153±0.106, P<0.05); the content of IL-10 was remarkably elevated (42.92±23.74 Vs 92.19±30.09,89.77±28.48, P<0.05). In TNBS group, the construction of tight junction was destroyed and the expressions of the related protein were lowed. After the treatment with the radix pulstillae alcohol extract and golden bifid, the condition of the damaged construction was better and the expressions of the related protein were rised (25.8±10.28 Vs 50.70±17.02,52.40±16.5, P<0.05).Conclusion:According to the research, radix pulstillae alcohol extract has the ability of protecting intestinal mucosal barrier. The mechanism is possibly related to up-regulate the tight junction protein between the epithelial cells of intestinal mucosal barrier through the mechanism of activating Occludin protein, which restrains endotoxin through the tight junction into the systemic circulation to reduce the level of TNF-a and to increase the production of IL-10 in order to anti-inflammatory effects.
