Objective:To research whether it could induce the expression and release of HMGB1 while TAP directly acts on the rat's pancreatic acini cell; to make a primary probe whether the relationship exists between the acute pancreatitis' TAP at early age and HMGB1 at advanced age in terms of cell field; as well as EP's inhibition on HMGB1.Method:The experiment was performed in 12 SD rats. Take out of pancreate, separate it into three groups and make comparison:comparison group (A), TAP group (B), and EP group(C). Group B and C were all added into TAP in different doses (1nmol/L,2nmol/L, and 3nmol/L);meanwhile,Group C was added into EP (28mmol/L) at the same time. Samples were respectively taken out from every group in random after 3h,6h,12h and 24h.Real-time quantitative reverse transcription polymerase chain reaction (RT-PCR) detected HMGB1mRNA expression; Western-blot probed HMGB1 protein expression. Analysis the correlation of HMGB1mRNA and protein expression with different TAP concentration and acting time.Results:As samples' TAP concentrations escalated in Group B and Group C, HMGB1mRNA and protein expression went up (p< 0.05), compared with those in Group A; compared with that in Group B, HMGB1mRNA and protein expression of Group C decreased (p<0.05); inside Group B, samples' HMGB1mRNA and protein went up as time went by and TAP concentrations-went up (p<0.05). Moreover, it increased dramatically at 12h (p<0.01). HMGB1 mRNA and protein expression have positive correlation with different TAP concentrations and acting time.Conclusion:TAP can directly induce the release of HMGB1 of pancreatic acini cell. Acute pancreatitis's TAP at early age has the norientation relationship with time and concentration. EP can curb the release of HMGB1 of acute pancreatitis.
