Objective:To investigate the expression of Hsp70 and NF-κB in the model of New Zealand rabbits lung ischemia reperfusion injury (IRI) and explore it's relationship with lung IRI.Methods:New Zealand rabbits lung IRI model were established by obstructing the left hilum of lung. Lung tissue samples were collected before ischemia,1h after ischemia/before reperfusion,30min,1h,2h,4h,6h post-reperfusion respectively. The mRNA expression of Hsp70 and NF-κB were determined by RT-PCR, and immunohistochemical method was used to detect their antibodies expression.Result:1.Hsp70 and NF-κB has statiscal significance between different times of IRI in mRNA and protein level(Hsp70,F=127.846,F=1532.780;NF-κB,F=641.180, F=328.111.P<0.01).2.The expression of the Hsp70 has the same trend in mRNA and protein level, it steps up after ischemia, advances after reperfusion, and reachs maximum 6h post-reperfusion. The expression between before reperfusion and 30min post-reperfusion has no statiscal difference(P>0.05),neither is before reperfusion and 1h post-reperfusion. There are statiscal differences between 1h,2h,6h post-reperfusion(P< 0.05).The expressions between 2h and 4h post-reperfusion,4h and 6h post-reperfusion has no statiscal difference(P>0.05).3.The expression of the NF-κB has the same trend in mRNA and protein level, it steps up after ischemia, advancs after reperfusion, reachs maximum 1h post-reperfusion, and then decreases.There is no statiscal difference between 30min and 2h post-reperfusion,30min and 4h post-reperfusion (P>0.05).The expression between 30min post-reperfusion and other times has statiscal difference(P< 0.05).2h post-reperfusion has no statiscal difference with 4h post-reperfusion (P>0.05), neither is 4h and 6h post-reperfusion (P>0.05).Expression 2h post-reperfusion is apparently different from that 6h post-reperfusion (P<0.05).4. The expression of Hsp70 is significantly negative correlation with the expression of NF-κB in rabbits after lung ischemia reperfusion.Conclusion:The expression of hsp70 increases after lung ischemia reperfusion; it's the endogenous protective molecules in lung ischemia reperfusion.The expression of NF-κB increases after lung ischemia reperfusion; it's involved in lung ischemia reperfusion injury acting as an important transduction factors.
