Expression Of VEGF-mRNA, UPA-mRNA, LRP, TS And ER In Breast Carcinoma As Well As Biological Significance

This paper detected the expression of vascular endothelial growthfactor (VEGF-mRNA), urokinase-type plasminogen activator (uPA-mRNA), lung resistance protein (LRP), thymidylate synthase (TS) andestrogen receptor (ER) in breast adenoma and carcinoma and explored thecorrelation between the two groups and their relationship with breastcancer biological characteristics. Therefore, it will provide a theoreticalbasis for the prognosis and guiding treatment of breast cancer patients.Specimens of breast tumor 128 cases (98 cases of breast carcinomaand 30 cases of breast adenoma) were examined by RNA in situhybridization method. All the patients were not treated by radiotherapy,chemo-treatment in preoperative. And the fresh specimens were fixed by4% paraformaldehyde for 4 hours, followed by gradient alcoholdehydration, xylene transparent, paraffin-soaked, and embedding, serialsectioning thickness of 7μm. Expression of VEGF-mRNA and uPA-mRNA were detected and analyzed by these kits combining laboratory conditions.At the same time, specimens of breast tumor 168 cases (130 cases of breastcarcinoma and 38 cases of breast adenoma) were detected byimmunohistochemistry technology SP method. All the patients were nottreated by radiotherapy, chemo-treatment in preoperative and the freshspecimens were fixed by 10% neutral formaldehyde for 24 hours, thendehydrated by alcohol, soaked in paraffin wax, embedded in paraffin wax,sliced for 4 um. Expression of LRP, TS and ER were detected and analyzedby these kits combining laboratory conditions. SPSS13.0 statisticalsoftware was used for statistical analysis, independent samplesχ2test and Fisher exact probability calculation was used to compare the rates.VEGF-mRNA, uPA-mRNA, and LRP, TS, ER correlations between thetwo groups, were detected by using non-hierarchical spearman correlationanalysis, with testing standard being P<0.05.The results are as follows:1. The positive rate of VEGF-mRNA was respectively 26.7%(8/30) and65.3%(64/98) in breast adenoma and breast carcinoma and there wassignificant difference between them (P<0.01); the positive rate ofVEGF-mRNA was 80.8%(42/52) in the axillary lymph nodes metastasisand 47.8%(22/46) in no the axillary lymph nodes metastasis in breastcarcinoma and there was significant difference between them (P<0.01).The expression of VEGF-mRNA had no significant difference withdifferent age groups, location and tumor size (P>0.05).The expression ofVEGF-mRNA in stageⅠis lower than inⅡandⅢstag and there wassignificant difference (P<0.05).2. The positive rate of uPA-mRNA was 20.0%(6/30) and 57.1%(56/98)respectively, there was significant difference between them (P<0.01); thepositive rate of uPA-mRNA was 73.1%(38/52) in the axillary lymph nodesmetastasis and 39.1%(18/46) in no the axillary lymph nodes metastasis inbreast carcinoma and there was significant difference between them(P<0.01). The expression of uPA-mRNAhad no significant difference withdifferent age stages group, location and tumor size (P>0.05);uPA-mRNAin stageⅠwas lower than inⅡstage and there was significant differencebetween them(P<0.05).3. There was significant difference between VEGF-mRNA anduPA-mRNA(P<0.05) with an obvious correlation coefficient (r=0.361).4. LRP, TS, and ER expressions in breast tumors positive substanceswere located in the cytoplasm and nuclei of tumor cells, showingbrown-yellow granules.5. The positive rate of LRP respectively was 21.1%(8/38) and 66.2%(86/130) in breast adenoma and breast carcinoma, (P<0.01); thepositive rate of LRP was 78.1%(57/73) in the axillary lymph nodesmetastasis and 50.9%(29/57) in no the axillary lymph nodes metastasis inbreast carcinoma and there was significant difference between them(P<0.01). The expression of LRP was no significant difference withdifferent age stages group, location, pathological styles and tumorsize(P>0.05);The expression of LRPin stage 0 was lower than inⅡandⅢstage, there was significant difference between them(P<0.05).6. The positive rate of TS respectively was 15.2%(6/38) and59.2%(77/130) in breast adenoma and breast carcinoma, (P<0.01); thepositive rate of TS was 71.2%(52/73) in the axillary lymph nodesmetastasis and 43.9%(25/57) in no the axillary lymph nodes metastasis inbreast carcinoma, there was significant difference between them (P<0.01);The expression of TS was no significant difference with different age stagegroups, location, pathological styles and tumor size (P>0.05), Theexpression of TS in stage 0 was lower than inⅡstage and there wassignificant difference between them(P<0.05).7. The positive rate of ER respectively was 36.8%(14/38) and57.5%(75/130) in breast adenoma and breast carcinoma and there wassignificant difference between them (P<0.05); the positive rate of ER was64.4%(47/73) in the axillary lymph nodes metastasis and 49.1%(28/57) inno the axillary lymph nodes metastasis in breast carcinoma, there was nosignificant difference between them (P>0.05). The expression of ER wasno significant difference with different age stage groups, location, clinicalstages and tumor size (P>0.05);The positive rate of ER was 66.7%(50/75)in invasive ductal carcinoma group , the positive rates of ER was 35% inother group(mainly the medullary carcinoma) and there was a significantdifference (P<0.05).8. There was no significant difference between LRP and ER (P>0.05)with the correlation coefficient (r=0.040);there was significant difference between TS and ER (P<0.05) with the correlation coefficient (r=0.178);there was significant difference between LRP and TS (P<0.01) with thecorrelation coefficient (r=0.332).Conclusion:1. The positive rate of VEGF-mRNA, uPA-mRNA, LRP,TS and ER inbreast carcinoma all were higher than those in breast adenoma, indicatingthat the five substances with the degree of benign, malignant tumorprogression.2. The expreessions of VEGF-mRNA and uPA-mRNA were closelyrelated to the axillary lymph nodes metastasis, they may participate in thedeterioration and metastasis of breast carcinoma; the expreessions ofVEGF-mRNA and uPA-mRNA were related to clinical stage of breastcancer, the higher stage, the higher the positive rate; the expression ofVEGF-mRNAand uPA-mRNAin breast cancer is unrelated to the patient'sage, location and tumor size.3. The expression of LRP, TS are closely related to the axillary lymphnode metastasis, they may take part in the deterioration and metastasis ofthe breast cancer; LRP, TS were related to clinical stages of breast cancer,the higher stage, the higher the positive rate; LRP, TS in the expression ofbreast cancer were unrelated to the patient's age, location and tumor size.4. The expression of ER was unrelated to axillary lymph node metastasis;the positive expression rate of ER in invasive ductal carcinoma was higherthan other group (mainly the medullary carcinoma), indicating that the lowexpression of ER in medullary carcinoma ; the expression of ER wasunrelated to the patient's age, location, tumor size and clinical stages inbreast cancer.5. The positive expression of VEG-FmRNAand uPA-mRNAbetween thetwo groups was significant (P<0.05, r = 0.361); the expression of LRP andTS between the two groups was significant (P<0.01, r=0.332); theexpression of LRP and ER between the two groups had no significant difference (P>0.05, r = 0.040); the expression of TS and ER between thetwo groups was significantly different (P<0.05, r=0.178); correlationcoefficient among the three groups were positive, there was synergy.Except the expression of LRPand ER between the two groups, all the othergroups were statistically significant, testing VEGF-mRNA, uPA-mRNA,LRP,TS, and ER will guide chemo-therapeutic drug selection.
