Ilex Cornuta is evergreen holly plant ,found in most parts of southern China. Soak fresh holly leaves, then dried, for tea with live blood, blood lipid and other effects. Ilex Cornuta leaves active chemical constituents, have been reported in the literature. Ilex Cornuta leaves extract and active chemical ingredient separation and purification of many, including traditional and modern technology. But active chemical constituents of the Root of Ilex Cornuta have few reports. In summary, this thesis the Root of Ilex Cornuta as raw material, grinding, soaking and other experiments on the pretreatment of raw materials, then the chemical composition on its activity for extraction, purification, identification and structural analysis of purity such as research, the following specific work :1.Rutin as reference substance, ethanol as extraction solvent, UV - spectrophotometry holly root extraction rate were measured. In the single factor experiment based on orthogonal experimental method to study the extraction time, extraction temperature, ethanol concentration, solid to liquid ratio of four factors on the Root of Ilex Cornuta extraction rate of flavonoids. The results showed that optimum conditions are: extraction temperature 80℃, solid to liquid ratio of 1:14 in ethanol (concentration 20%), extraction time was 3h.2.Purification by silica gel column chromatography, respectively, with petroleum ether, ethyl acetate, chloroform, butanol followed by elution by a pale yellow needle-like pure substance, the substance is soluble in glacial acetic acid, insoluble in water. After IR, NMR, MS and characterized to determine the substance is quercetin, its structure is shown below.3.In this thesis, the Root of Ilex Cornuta as raw material, simplifying the extraction and separation processes, extraction and isolated by a kind of flavonoid quercetin active chemical substances, for the Root of Ilex Cornuta separation and extraction of bioactive components was provided with some experimental foundation.
