Investigation of antivasospasmic effect of Ginkgo biloba extract in experimental subarachnoid hemorrhage of ratsObjective:to study the influence of Ginkgo biloba extract on cerebral vasospasm after experimental subarachnoid hemorrhage of rats, and to observe the changes of cerebrovascular wall including the thickness and the expression of platelet activating factor, thus to confer whether Ginkgo biloba extract can effectively reverse the brain damage caused by cerebral vasospasm after experimental subarachnoid hemorrhage and find out the role of PAF in this process.Methods:A model of vasospasm was constructed by injecting self-femoral artery blood into cisterna magna. thirty selected Sprague-Dawley rats were randomly divided into five groups:sham-operated group were the untreated control group, saline group, vasospasm was induced in SAH group, Ginkgo biloba extract Intervention group and Ginkgo biloba extract preprocessing group. Score of animal neurological behavior at 3d,7d were performed after modeling. Then removed brain pontine basilar artery at 7d, observed the sections of blood vessels spasm and cerebrovascular wall morphology changes in biopsy, and used immunohistochemical method to determine the expression changes of PAF in vascular wall. All of the data were analyzed using the Statistical Package for the Social Sciences for Windows version 18.0.Results:the animal neurobehavioral score was the highest at the third day of SAH group, compared with Ginkgo biloba extract Intervention group and Ginkgo biloba extract preprocessing group, there was significant statistical difference (P<0.05), In SAH group, signs of inflammation were observed, as well as marked increase in the thickness of the vessel wall, also there was significant statistical difference compared with Intervention group and preprocessing group (P<0.05), but no statistical difference between Intervention group and preprocessing group (P>0.05). The SAH group showed the strongest PAF expression at the time of 7d followed by the Intervention group and preprocessing group, The saline group and the sham group had no significant expression of PAF.Conclusion:1. Local inflammatory reaction in vascular wall may be related to PAF which performed a important role in chronic cerebral vasospasm after subarachnoid hemorrhage.2. Ginkgo extract may inhibit the expression of PAF to effectively alleviate the severity of chronic vasospasm induced by subarachnoid hemorrhage.3.there was no significant difference on the prognosis of rats suffering from cerebral vasospasm after subarachnoid hemorrhage between Prophylactic use of Ginkgo biloba extract and use of Ginkgo biloba extract after SAH.
