Background:Lung cancer is the most common and heavy burden disease in the world.The invasion and metastasis is one of the most important factors that influence the treatment and prognosis of patients who have lung cancer. Metastasis is a multi-stage process involving cancer cell motility,intravasation,transit in the blood or lymph,extravasation and growth at a new site.In current,much research indicates that there are many factors influence metastasis that such as EMC(extracellar matrix), CAM (cell adhesion molecule), proteolytic ferment, VEGF (Vascular endothelial growth factor), metastasis related gene and miRNAs and so on.MicroRNA(miRNA) is a kind of small noncoding single-strand RNA and it is about 19-24 nucleotide (nt).MiRNAs have been widely identified in plants and animals.They play a important role in developments such as cell differentiation, proliferation, apoptosis, metabolism, infection, especially generation and metastasis of tumour. Metastasis is a important character of tumour,and scientists in the world are paying close attentions to the relationship of miRNAs and metastasis.Many studies report that miRNAs play an important role in the metastasis of breast cancer, liver cancer, gastric cancer and thyroid cancer and so on.The study of metastasis related microRNAs in lung cancer is very little.In this study, we plan to screen and search the miRNAs that involed in metastasis of lung cancer.Objective:Screen and search the miRNA,which differentially express in 95d cell line and 95c cell line by using microarray.Methods:The 95d cell line and 95c cell line were cultivated in routine method,Total RNA of 95d cell and 95c cell was isolated by Trizol.MiRNAs were isolated using PEG and were labeled by RNA joining enzyme.Then the miRNAs were hybridized on miRNA Microarray and scanned.Using LuxScan 3.0 image analysis software analyze and original image signal was diverted into digit signal.The differentially espressed miRNAs were screened and identified though comparison between the two chips.Results:The results of miRNA microarray show that 24 miRNAs expressed differentially between 95d and 95c cells. hsa-miR-138-2*, hsa-miR-487a, hsa-miR-151-5p, hsa-miR-125a-3p, hsa-miR-220b, hsa-miR-1229, hsa-miR-125a-5p, hsa-miR-1290, hsa-miR-18a mmu-miR-1195, hsa-miR-487b, hsa-miR-652, hsa-miR-1285 mmu-miR-700, hsa-miR-663, hsa-let-7g, hsa-miR-222, and predicted miRNAs MIR232,MIR196,MIR237,MIR189,MIR191 were up-regulated; mmu-miR-220,hsa-miR-5481 were down-regulated.Conclusions:1.MiRNAs were differentially expressed between 95d and 95c cells. 2.The 22 up-regulated miRNAs in 95d cell may promote lung cancer metastasis,and the 2 down-regulated miRNAs may inhibit lung cancer metastasis.
